تجزیه ارتباط رگرسیونی صفات مرتبط با عملکرد با نشانگرهای مولکولی RAPD در پسته (P. vera L)
با بکارگیری نشانگرهای مولکولی، اصلاح گیاهان با سرعت و سهولت بیشتری انجام میگیرد و انتخاب والدین برای تلاقی های بعدی در برنامههای اصلاحی با اطمینان بیشتری انجام میگیرد. در دسترس بودن تعداد بسیار زیادی از نشانگرها و صفات مورفولوژیکی میتواند به مطالعه تجزیه رگرسیونی بین این نشانگرها و صفات مورفولوژیکی کمک نماید. در این تحقیق، ارتباط صفات مرتبط با عملکرد در 20 ژنوتیپ پسته با استفاده از 15 آغازگر RAPD مورد بررسی قرار گرفت. در نهایت 11 آغازگر چندشکلی نشان دادند و مجموعاً 56 قطعه (لوکوس) را تکثیر کردند که از این بین 36 قطعه (29/64 درصد) چندشکلی را با میانگین 09/5 الل به ازای هر پرایمر نشان دادند و میزان این چندشکلی از حداقل 25 درصد برای آغازگر AJ05 تا حداکثر 5/87 درصد برای آغازگر OPAD02 متغیر بود. میانگین محتوای اطلاعاتی حاصل از چند شکلی برای جایگاهها 23/0 و از 095/0 (AJ05 و OPAD14) تا 39/0 (OPC05) متغیر بود. برای شناسایی نشانگرهای مثبت مرتبط با صفات اجزای عملکرد، تجزیه رگرسیون گام به گام بین دادههای مولکولی به عنوان متغیرهای مستقل و صفات مورد مطالعه به عنوان متغیرهای وابسته انجام گرفت. نوزده قطعه RAPD با شش صفت مرتبط با عملکرد ارتباط داشتند. بعضی از نشانگرهای RAPD با بیشتر از یک صفت در تجزیه رگرسیون چندگانه ارتباط داشت که میتواند به خاطر اثر پلیوتروپیک مکانهای صفات کمی بر روی صفات مختلف یا پیوستگی ژنهای مختلف باشد. برای درک این موضوع تهیه نسلهای در حال تفرق و نقشههای پیوستگی ضروری میباشد. همچنین این نتایج میتواند در برنامههای اصلاحی انتخاب به کمک نشانگر هنگامی که هیچ اطلاعات ژنتیکی در دسترس نیست، مفید باشد.

بررسی سرعت رشد میسلیوم و عملکرد جدایه های مختلف قارچ خوراکی دکمه ای سفید (Agaricus bisporus) در ایران
در بین قارچهای خوراکی، قارچ خوراکی تکمهای سفید، رایجترین قارچی است که در سراسر جهان کشت میشود. به منظور بررسی سرعت رشد میسیلیوم و عملکرد جدایههای قارچ خوراکی دکمهای سفید در محیط کشت جامد، اسپاون و کمپوست، پژوهشی در پژوهشکده قارچ دانشکده کشاورزی دانشگاه فردوسی مشهد، در سال 1393 انجام شد. هجده جدایه قارچ خوراکی دکمهای سفید از لحاظ سرعت رشد میسیلیوم روی محیط-های کشت پوتیتو دکستروز آگار (PDA)، عصاره کمپوست (CYM) و کمپوست، کلاس رشدی و تیپ رشدی میسلیوم و عملکرد مورد مقایسه قرار گرفتند. نتایج نشان داد که تفاوت بسیار معنیداری بین جدایههای قارچ خوراکی دکمهای از نظر سرعت رشد میسیلیوم در محیطهای کشت و میزان عملکرد وجود دارد. در محیط کشت PDA، بیشترین سرعت رشد به میزان 9/1 میلیمتر در روز و قطر نهایی پرگنه 1/8 سانتیمتر، متعلق به جدایه 2200 بود. همچنین، این جدایه از لحاظ سرعت رشد میسیلیوم در محیط CYM و پوشاندن سطح اسپاون و کمپوست نیز جزء جدایههای سریع بود و به همراه جدایه A15a از لحاظ عملکرد نیز از جمله جدایههای با عملکرد زیاد بودند (A15a و 2200 به ترتیب 1/22 و 4/19 کیلوگرم در متر مربع). همچنین، بر اساس نتایج به دست آمده، سرعت رشد میسیلیوم با عملکرد همبستگی مثبت و معنیداری داشت و جدایههای با سرعت رشد میسیلیوم بیشتر عملکرد بیشتری تولید کردند. طبق نتایج به دست آمده، همبستگی مثبت و معنیداری بین سرعت رشد میسیلیوم در محیط کشت CYM و سرعت پوشاندن سطح کمپوست توسط میسیلیوم مشاهده شد، که میتواند به عنوان محیط کشت مناسب برای مقایسه سرعت رشد میسیلیوم در محیط آزمایشگاهی مورد استفاده قرار گیرد.

اثرات مصرف اسید جیبرلیک بر بیدانه کردن و برخی صفات کمی و کیفی میوه سه رقم انگور دانه دار ایرانی
به منظور مطالعه اثرات اسید جیبرلیک در دو زمان 7 و 14 روز قبل از شروع گلدهی بر القاء بیدانگی و بعضی صفات کمی و کیفی خوشه و حبه سه رقم انگور دانهدار ایرانی شامل قزل اوزوم، ریش بابا قرمز و خلیلی قرمز، پژوهشی به صورت آزمایش فاکتوریل در قالب طرح بلوکهای کامل تصادفی در پنج تکرار اجرا شد. نتایج نشان داد بوتههای تیمار شده با جیبرلین در مقایسه با شاهد از لحاظ درصد جوانهزنی دانه گرده، درصد تشکیل میوه، طول و وزن تر چوب خوشه، تعداد شات بری و حبههای بیدانه در خوشه، وزن حبههای بیدانه، طول دم حبه و TSS اختلاف معنی دار نشان دادند. جیبرلین باعث کاهش جوانهزنی دانه گرده ارقام مورد بررسی شد. تیمار جیبرلین درصد تشکیل میوه را افزایش داد اما واکنش ارقام متفاوت بود. اسید جیبرلیک موجب افزایش طول محور خوشه انگور شد و این افزایش در رقم ریش بابا و قزل اوزوم بیشتر از خلیلی بود. نتایج نشان داد که هورمون جیبرلین باعث ایجاد بیدانگی در حبهها شد و اثر آن روی رقم ریش بابا قرمز بیشتر از قزل اوزوم و خلیلی قرمز بود. در انگور قزل اوزوم تیمار شده با جیبرلین در مرحله 7 روز قبل از گلدهی وزینترین حبههای بیدانه بدست آمد، گرچه تعداد حبههای بیدانه این رقم کمتر از ارقام دیگر بود. جیبرلین موجب افزایش طول دم حبهها شد. بیشترین طول دم حبه مربوط به رقم قزل اوزوم بود که در 14 روز قبل از گلدهی با جیبرلین تیمار شده بود. درصد مواد جامد محلول (TSS) در تیمارهای غوطه ورسازی شده با جیبرلین بیشتر از تیمارهای شاهد بود. مصرف قبل از شروع گلدهی هورمون جیبرلین با غلظت 100 میلیگرم در لیتر در هر سه رقم مورد بررسی باعث تولید حبههای بیدانه ریز شد که در صورت تکرار مصرف هورمون جیبرلین با غلظت 20 تا 40 پیپیام در مرحله بعد از تشکیل میوه میتوان حبههای بیدانه با اندازه قابل قبول تولید نمود.

بررسی تاثیر تنش خشکی بر فتوسنتز، درصد اسانس و عملکرد اسانس بابونه آلمانی (Matricaria recutita L.) رقم پرسو و کاسنی (Cichorium intybus L) توده محلی خوزستان در شرایط آب و هوایی شمال خوزستان
این تحقیق به منظور بررسی واکنش فتوسنتز و عملکرد اسانس گیاهان دارویی کاسنی توده محلی خوزستان و بابونه آلمانی رقم پرسو به تنش خشکی در شهرستان شوشتر (دانشکده کشاورزی دانشگاه آزاد اسلامی واحد شوشتر) در سال زراعی 90-89 به صورت دو آزمایش جداگانه انجام شد. آزمایشها در قالب طرح بلوک کامل تصادفی در چهار تکرار اجرا شد و تیمارهای تنش خشکی عبارت بودند از شاهد (حدود 90 درصد ظرفیت زراعی خاک)، تنش ملایم (حدود 75 درصد ظرفیت زراعی خاک) و تنش شدید (حدود 55 درصد ظرفیت زراعی خاک). صفات مورد بررسی در این تحقیق عبارت بود از ارتفاع بوته، وزن خشک برگ (کاسنی)، وزن خشک گل (بابونه)، غلظت کلروفیل a و b، فتوسنتز، درصد اسانس، عملکرد اسانس و اجزای تشکیل دهنده اسانس. نتایج آزمایش نشان داد تنش خشکی سبب کاهش فتوسنتز، غلظت کلروفیل a، وزن خشک و ارتفاع بوته بابونه و کاسنی شد اما درصد اسانس این گیاهان افزایش یافت. کمترین وزن خشک گل بابونه و وزن خشک اندام هوایی کاسنی تحت تاثیر تنش خشکی شدید به میزان 47 و 195 گرم بر متر مربع دیده شد در حالیکه بیشترین میزان عملکرد اسانس بابونه آلمانی (6/56 گرم در متر مربع) و کاسنی (9/225 گرم در مترمربع) تحت تاثیر تیمار تنش خشکی ملایم به دست آمد. در شرایط تنش خشکی ملایم بیشترین ترکیبات تشکیل دهنده اسانس بابونه کامازولن، آلفا پنین، آلفا بیسابول و بتا پنین بود در حالیکه بیشترین ترکیبات تشکیل دهنده اسانس کاسنی در شرایط تنش ملایم خشکی کامفور، آلفا پنین، بتا پنین و سابینین بود. به طور کلی نتایج نشان داد تنش خشکی ملایم به دلیل افزایش عملکرد اسانس علی رغم کاهش معنیدار وزن خشک اندام اقتصادی سبب افزایش عملکرد اسانس گیاهان مورد بررسی شد.

بررسی ترکیبپذیری عمومی، خصوصی و هتروزیس صفات رویشی تعدادی از لاین های خیار
ارزیابی ترکیبپذیری عمومی و خصوصی، هتروزیس و نحوه عمل ژنها برای صفات طول بوته، تعداد شاخه فرعی و طول بوته تا اولین میوه در قالب آزمایش دیآلل ناقص 7×7 با استفاده از روش دوم و چهارم در مدل ثابت گریفینگ برای تعدادی از لاینهای خیار در سال 1393 صورت گرفت. میانگین مربعات ترکیبپذیری عمومی فقط برای صفت طول بوته تا اولین میوه معنیدار بود ولی میانگین مربعات ترکیبپذیری خصوصی برای تمامی صفات معنیدار شد که مبین اهمیت بیشتر اثرات غالبیت ژنها در توارث این صفات در جمعیت مورد مطالعه میباشد. به نظر میرسد صفت طول بوته تا اولین میوه با توجه به ترکیب پذیری عمومی بالا توسط عمل افزایشی ژنها کنترل میشود.

بررسی تحمل به شوری دانهال های ارقام تجاری و ژنوتیپ های محلی پسته (Pistacia vera L.) منطقه رفسنجان در شرایط کنترل شده
در ایران عمده اراضی زیرکشت پسته در حاشیه کویر قرار دارد و یکی از مشکلات عمده این اراضی شوری خاک و آب آبیاری است که رشد و عملکرد گیاه را تحت تاثیر قرار میدهد. در این تحقیق اثرات شوری روی خصوصیات رشدی و مقدار عناصر معدنی دانهالهای 10 رقم پسته (اکبری، احمدآقایی، کله قوچی، فندقی، بادامی، ابراهیمی، سیف الدینی و ژنوتیپهای G1، G2 وG3) به صورت کرتهای خرد شده در قالب طرح بلوکهای کامل تصادفی در سه تکرار مطالعه شد. عامل اصلی مقادیرشوری آب آبیاری (6/0، 15 و 30 دسی زیمنس بر متر) با استفاده از نمک کلرور سدیم و عامل فرعی ارقام پسته بود. بذور پس از جوانهزنی در آزمایشگاه به داخل گلدان در گلخانه منتقل شدند و بعد از مرحله سه برگی، تیمارهای شوری به مدت چهارماه اعمال گردید. نتایج نشان داد با افزایش شوری وزن خشک ساقه و ریشه، طول نهال، وزن خشک برگ، تعداد برگ، سطح برگ و قطر نهال کاهش داشت. با افزایش شوری، میزان عناصر سدیم و کلسیم در ریشه، ساقه و برگ افزایش و نسبت پتاسیم به سدیم در این اندامها کاهش یافت. ارقام فندقی و کله قوچی بیشترین و ژنوتیپهایG1 و G3 دارای کمترین میزان سدیم برگ بودند. ژنوتیپ G2 بیشترین میزان (3 درصد) و رقم کله قوچی کمترین میزان (4/1 درصد) پتاسیم برگ را دارا بودند. به طورکلی ژنوتیپهای محلی پسته ( G1، G2 و G3) جذب و انتقال سدیم کمتری را به برگ انجام دادند، به طوریکه ژنوتیپ G3 دارای بیشترین سدیم ریشه و کمترین سدیم ساقه و برگ بود. ژنوتیپG2 و رقم احمدآقایی به ترتیب با مقادیر 8/7 و 3/6 بیشترین نسبت پتاسیم به سدیم در برگ و رقم کلهقوچی کمترین نسبت (9/3) را داشت. به نظرمیرسد ارقام کله قوچی، فندقی و اکبری در مراحل اولیه رشد نسبتا حساس و ژنوتیپهای G1، G2 وG3 و رقم احمدآقایی نسبتا متحمل به شوری باشند. ارقام بادامی، ابراهیمی و سیف الدینی از نظر تحمل به شوری در حد متوسط بودند.

تاثیر اسید سالیسیلیک و سولفات منیزیم کلاته بر تخصیص مواد در قسمتهای رویشی و زایشی در گلابی رقم لوئیزبون آلوده به بیماری آتشک
توزیع مواد فتوسنتزی بین قسمتهای رویشی و زایشی تحت تاثیر عوامل محیطی و وضعیت تغذیه گیاه قرار میگیرد (20). هر یک از قسمت-های رویشی و زایشی گیاه میتوانند به عنوان مخزن (Sink)، بخشی از مواد پرورده فتوسنتزی را به خود اختصاص دهد. بنابراین تغذیه با مواد مختلف تیماری میتواند توزیع مواد فتوسنتزی بین قسمتهای رویشی و زایشی گیاه را تحت تاثیر قرار داده و تغییراتی در تخصیص مواد پرورده فتوسنتزی بوجود آورد. از آنجائیکه بررسی تسهیم مواد پرورده فتوسنتزی مبحث بسیار پیچیدهای بوده و تفسیر اثرات تیمارها بر تسهیم مواد در گیاهان باغی دشوار است، لذا این بررسی با مقایسه تاثیر تیمارهای اسید سالیسیلیک و سولفات منیزیم کلاته بر تخصیص مواد پرورده به پارامترهای رویشی و زایشی، کاربرد خارجی تیمارهای تغذیهای را بر رشد اندامهای مختلف هدف قرار داده است. در تحقیق حاضر اندازهگیری پارامترهای رشد رویشی (وزن برگ، سطح برگ، محتوای نسبی آب برگ، رشد شاخه سال جاری و غیره) و زایشی (نسبت طول به قطر و چگالی میوه و غیره) در شرایط اعمال تیمارهای اسید سالیسیلیک و سولفات منیزیم کلاته با هدف بررسی تاثیر کاربرد خارجی مواد هورمونی و معدنی بر پارامترهای مزبور در شرایط آلودگی طبیعی آتشک و در راستای نیازهای تولید صورت گرفت. سطوح تیماری اسید سالیسیلیک صفر، 1/0 و 5/0 گرم در هزار و سولفات منیزیم کلاته صفر، 5/0 و 7/0 گرم در هزار بود. نتایج ارزیابیها نشان داد که ترکیبات تیماری تخصیص مواد پرورده فتوسنتزی را تغییر داده و بین ترکیبات مختلف تیماری از این نظر اختلاف معنیداری وجود داشت (01/0p

بررسی تنوع مورفولوژیکی و فیزیولوژیکی برخی از تودههای فلفل شیرین (.Capsicum annuum L)
فلفل از جمله مهمترین سبزیجات به شمار میرود که علاوه بر مصارف غذایی، دارای مصارف دارویی نیز میباشد. به منظور مطالعهی تنوع ژنتیکی فلفل شیرین از نظر صفات مورفولوژیک و فیزیولوژیک، 42 توده فلفل در قالب طرح لاتیس مستطیل در 3 تکرار در ایستگاه تحقیقات کشاورزی ساعتلوی ارومیه در سال 1392 مورد ارزیابی قرار گرفت. نتایج نشان داد بین تودهها برای کل صفات به جزء صفات میزان اسپد، میزان فتوسنتز و pH اختلاف آماری معنیداری وجود دارد. بیشترین همبستگی فنوتیپی مثبت (958/0) و بیشترین همبستگی ژنتیکی مثبت (994/0) بین صفات وزن خشک میوه و وزن تر میوه و بیشترین همبستگی ژنتیکی منفی (587/0) بین صفات کانوپی گیاه و عرض میوه مشاهده شد. بر اساس جدول مقایسه میانگین ژنوتیپهای ارومیه، لردگان و اورفای ترکیه دارای عملکرد بیشتری نسبت به سایر ژنوتیپها بودند. براساس نتایج به دست آمده صفات طول میوه، عرض میوه، ضخامت دیواره میوه، عملکرد، وزن تر میوه و وزن خشک میوه دارای توارث پذیری بالای در بین صفات مورد بررسی بودند. با استفاده از تجزیهی خوشهای به روش وارد، تودههای مورد بررسی بر اساس صفات مورفولوژیکی و فیزیولوژیکی در 6 گروه متفاوت قرار گرفتند و بر این اساس بیشترین فاصله بین دو خوشهی چهارم و پنجم مشاهده شد.

تاثیر حلالهای مختلف بر میزان ترکیبات فنلی و فعالیت آنتی اکسیدانی میوه عناب (Ziziphus jujube Miller)
این تحقیق به منظور بررسی اثر تیمار حلالهای متانول، اتانول، استون (50، 90 و 100 درصد) و آب مقطر بر روی میزان ترکیبات آنتی اکسیدانی (فنل کل، تانن ها، فلاونوئیدها، آنتوسیانین ها) میوه عناب (Ziziphus jujube Miller)، انجام شد. فعالیت آنتی اکسیدانی هر یک از عصارهها نیز به روشهای DPPH وFRAP مورد ارزیابی قرار گرفتند. نتایج نشان داد که استون 50 درصد در استخراج ترکیبات فنل و فلاونوئید کل به ترتیب با مقادیر (20/798 میلی گرم گالیک اسید در 100 گرم ماده خشک) و (73/1871 میلی گرم کوئرستین در 100 گرم ماده خشک)، بهترین عملکرد را در میان سایر حلالها داشته است. استخراج آنتوسیانین، با متانول 50 درصد (31/6 میلی گرم سیانیدین 3- گلوکوزید در 100 گرم ماده خشک) و استخراج تانن با متانول 100 درصد (4/669 میلی گرم کاتکین در 100 گرم ماده خشک)، دارای بالاترین بازده استخراج بوده است. در تمامی موارد آب از حداقل قابلیت استخراج متابولیتهای ثانویه در بین سایر حلالها برخوردار بود. در بررسی خاصیت آنتی اکسیدانی میوه عناب به دو روش DPPH و FRAP مشخص گردید که خاصیت آنتی اکسیدانی آنها مستقیما تحت تاثیر مقادیر ترکیبات فنلی قرار گرفته است. در میان حلالهای مورد استفاده، استون 50 درصد بهترین بازده را در روش DPPH ( 02/29=IC50) و روش FRAP (11/5 میلی مول آهن II در 100 گرم ماده خشک) نسبت به سایر حلالها داشت. بر اساس نتایج میتوان گفت درصد مهار کنندگی DPPH و FRAP با کاهش غلظت حلالهای متانول و استون رابطه مستقیم دارد. نتایج این تحقیق نشان داد که تفاوت در نوع، خلوص و قطبیت حلالها و تفاوت در جزئیات روش استخراج عصاره (مانند زمان استخراج، سرعت اختلاط، نسبت میزان پودر به حلال و اندازه ذرات پودر) بر میزان استخراج متابولیتهای ثانویه تاثیر میگذارند.

بررسی خصوصیات جوانه زنی توده های گیاه دارویی شنبلیله (Trigonella foenum-graecum L.) تحت تأثیر تنش شوری و خشکی
مرحله جوانهزنی گیاهان یکی از مراحل مهم در طول دوره رشد آنها میباشد که اغلب تحت تأثیر تنشهای محیطی به ویژه شوری و خشکی قرار میگیرد. به منظور بررسی اثر تنشهای شوری و خشکی دو آزمایش جداگانه به صورت فاکتوریل در قالب طرح کاملاً تصادفی با سه تکرار بر روی بذر تودههای بومی شنبلیله آمل، تبریز، ساری و مشهد در آزمایشگاه تحصیلات تکمیلی گروه علوم باغبانی دانشگاه فردوسی مشهد در سال 1393 اجرا شد. تیمارهای شوری به ترتیب (صفر، 60، 120 و 180 میلیمولار نمک کلرید سدیم) و تیمارهای خشکی (شاهد، 3-، 6- و 9- بار پلی اتیلن گلایکول 6000) بودند. نتایج نشان داد که با افزایش سطوح شوری و منفی تر شدن پتانسیل اسمزی درصد و سرعت جوانهزنی، طول ریشهچه، طول ساقهچه و وزن خشک گیاهچه به طور معنیداری (01/0P≤) در کلیهی تودههای مورد بررسی نسبت به شاهد کاهش یافت. میزان کاهش این صفات در تنش شوری نسبت به تنش خشکی بیشتر بود که نشان دهنده حساسیت بیشتر تودههای شنبلیله به تنش شوری نسبت به تنش خشکی است. بین تودهها، توده-های مشهد و آمل متحملترین و تودههای تبریز و ساری حساسترین تودهها به شوری و خشکی بودند. بر اساس نتایج پژوهش حاضر، به نظر میرسد بتوان از تودههای مشهد و آمل برای اصلاح تودههایی با پتانسیل عملکرد زیاد در مناطق شور و خشک استفاده کرد.

تأثیر محلولپاشی با کلرور کلسیم بر عارضة پوسیدگی گلگاه در دو رقم هندوانه (.Citrullus lanatus (Thunb) Matsum&Nakai)
بهمنظور بررسی اثر محلولپاشی با کلرور کلسیم بر عارضة پوسیدگی گلگاه در دو رقم هندوانه آزمایشی بهصورت فاکتوریل در قالب طرح پایه بلوکهای کامل تصادفی در سه تکرار بر روی ارقام چارلستونگری و چارلی درایستگاه تحقیقات کشاورزی گنبد ودر سال زراعی 1387 اجرا شد. عوامل آزمایش شامل دو رقم هندوانه (چارلستونگری و چارلی) و چهار سطح محلولپاشی کلسیم (0، 3، 5 و 8 در هزار کلرور کلسیم) بود. صفات اندازهگیری شده شامل: متوسط وزن میوه، متوسط طول میوه، طول قسمت پوسیده میوه، بریکس، ضخامت پوست، عملکرد میوه، تعداد میوه آلوده و تعداد میوه سالم بود. نتایج تجزیه برگ و میوه نشان داد که محلولپاشی با کلرور کلسیم سبب افزایش کلسیم (از 24/. درصد به 3/0 درصد) در بافت آنها شد. اثر محلولپاشی سطوح مختلف کلرور کلسیم بر متوسط طول میوه، طول قسمت پوسیده، عملکرد میوه و تعداد میوه معنیدار نبود، درحالیکه محلولپاشی با کلرور کلسیم تعداد میوههای آلوده را بهطورمعنیداری کاهش داد، بهطوریکه تیمارهای صفر در هزار و 8 در هزاربه ترتیب 1667 و 588 میوه آلوده در هکتار داشتند.

Gemmifera) آزمایشی با شش سطح سلنیوم (0، 2، 4، 8، 16و 32 میلیگرم در لیتر از منبع سلنات سدیم) در قالب طرح کاملا تصادفی با چهار تکرار در شرایط گلخانهای انجام شد. نتایج نشان داد که خصوصیات رویشی گیاهان با افزایش غلظت سلنیوم تا 8 میلیگرم در لیتر افزایش و بعد از آن کاهش یافت. عملکرد گیاه نیز بطور معنیداری (P≤0.01) تحت تاثیر تیمارهای سلنیومی قرار گرفت، بطوریکه با افزایش غلظت سلنیوم در محلول غذایی از 0 تا 8 میلیگرم در لیتر عملکرد افزایش و سپس کاهش یافت. حداکثر عملکرد در غلظت 8 میلیگرم در لیتر مشاهده شد که نسبت به شاهد40 درصد افزایش عملکرد داشت. بنظر میرسد افزایش سطح برگ یکی از دلایل عمده در افزایش عملکرد در اثر افزودن سلنیوم به محلول غذایی باشد. بر اساس یافتههای این تحقیق سلنیوم را میتوان حداکثر تا غلظت 8 میلیگرم در لیتر به منظور بهبود رشد و نمو گیاه به محلولهای غذایی اضافه نمود.

بهینه سازی کالوس زایی و باززایی دو توده بومی گیاه دارویی شنبلیله (Trigonella foenum– graecum) در شرایط درون شیشه ای
شنبلیله با نام علمی Trigonella foenum– graecum گیاهی یکساله از تیره بقولات است. ریشه، برگ و بذر آن دارای ترکیبات دارویی مهمی میباشد. تحقیق حاضر به منظور تعیین مناسبترین غلظت تنظیم کنندههای رشد گیاهی برای تولید گیاهچههای درونشیشهای با استفاده از ریزنمونههای کوتیلدون و هیپوکوتیل دو ژنوتیپ ایرانی (اردستانی و نیشابوری) شنبلیله انجام گرفت. ریزنمونهها در محیط کشت MS حاوی تنظیم کنندههای رشد گیاهی IBAو TDZ جهت القای کالوس و باززایی مستقیم کشت گردیدند. در این آزمایش از دو ترکیب TDZ + IBAاستفاده شد. در ترکیب اول، IBA دارای 4 سطح (0، 1/0، 3/0، 5/0 میلیگرم در لیتر) و TDZ دارای 5 سطح (0، 2/0، 4/0، 6/0، 8/0 میلیگرم در لیتر) و در ترکیب دوم، IBA دارای 4 سطح (0، 05/0، 1/0، 15/0 میلیگرم در لیتر) و TDZ دارای 7 سطح (0، 2/0، 25/0، 3/0، 35/0، 4/0، 45/0 میلیگرم در لیتر) بودند. آزمایشها بصورت فاکتوریل در قالب طرح کاملاً تصادفی با 4 تکرار انجام گرفت. نتایج آزمایش نشان داد که ریزنمونههای کوتیلدون و هیپوکوتیل ژنوتیپهای اردستانی و نیشابوری کالوسزایی کردند، در حالیکه باززایی مستقیم تنها در ریزنمونههای هیپوکوتیل ژنوتیپها مشاهده شد. بیشترین درصد کالوسزایی ریزنمونههای کوتیلدون (75 درصد) در ژنوتیپ نیشابوری و در محیط کشت MS حاوی IBAmg l-1 5/0 + TDZmg l-1 4/0 بدست آمد. همچنین بیشترین درصد کالوسزایی ریزنمونههای هیپوکوتیل (75 درصد) در ژنوتیپ اردستانی مشاهده شد که در محیط کشت MS حاوی IBAmg l-1 5/0 + TDZmg l-1 8/0 بدست آمد. بیشترین درصد باززایی مستقیم (5/37 درصد) در ریزنمونههای هیپوکوتیل ژنوتیپ نیشابوری در محیط کشت MS حاوی IBAmg l-1 05/0 +TDZmg l-1 35/0 حاصل شد.

اثر اصلاح کننده های خاک و کودهای بیولوژیک بر خصوصیات مورفولوژیک و عملکرد گاوزبان ایرانی (Echium amoenum)
بهمنظور بررسی اثر اصلاحکنندههای خاک و کودهای بیولوژیک مختلف بر خصوصیات مورفولوژیکی و عملکرد گیاه دارویی گاوزبان ایرانی (Echium amoenum)، آزمایشی در سالهای زراعی 92-1390 در دانشگاه فردوسی مشهد در بهصورت طرح بلوکهای کامل تصادفی با سه تکرار انجام شد. تیمارهای آزمایش شامل هفت نوع اصلاحکنندهی خاک و کود بیولوژیک مختلف از جمله: 1- اسید هیومیک، 2- اسید فولویک، 3- نیتروکسین (حاوی باکتریهای Azotobacter spp. و Azospirillum spp.)، 4- بیوفسفر (حاوی باکتریهای Bacillus sp. و Pseudomonas sp.)، 5- بیوسولفور (Thiobacillus spp.)، 6- میکوریزا (Glomus mosseae)، و 7- میکوریزا (Glomus intraradices) و عدم استفاده از کود بهعنوان تیمار شاهد بودند. نتایج آزمایش نشان داد که هر دو گونه میکوریزای مورد مطالعه منجر به افزایش عملکرد گل در مقایسه با شاهد شدند، بهطوریکه عملکرد گل در تیمارهای Glomus mosseae و Glomus intraradices بهترتیب 24 و 11 درصد بیشتر از شاهد بود. تمامی نهادههای اکولوژیک مورد مطالعه سبب افزایش تعداد چرخهی گل در بوته در مقایسه با شاهد شدند، ولی اثر کود بیولوژیک بیوفسفر از این نظر بهطور بارزتری نمایان شد، به-طوریکه تعداد چرخهی گل در بوته از 342 چرخه در بوتههای تحتتیمار شاهد به 1322 چرخه در گیاهان تحتتیمار بیوفسفر افزایش یافت. بیشترین مقدار عملکرد دانه، عملکرد مادهی خشک، وزن دانه در بوته و تعداد دانه در بوته در تیمار اسید هیومیک مشاهده شد، بهطوریکه در نتیجهی کاربرد اسید هیومیک عملکرد دانه، عملکرد مادهی خشک، وزن دانه در بوته و تعداد دانه در بوته بهترتیب 82، 66، 63 و 66 درصد نسبت به شاهد افزایش یافتند.

تاثیر زمان برداشت و نوع بافت میوه بر میزان هسپریدین موجود در ارقام میوه لیموترش
فلاونوئیدها از مواد غذایی هستند که خود بخشی از ترکیبات معطر پلیفنلها میباشند. هسپریدین یکی از فلاونوئیدها میباشد که کاربرد زیادی در درمان بسیاری از بیماریها بویژه واریس پا دارد. هدف از این تحقیق تاثیر نوع بافت و زمان برداشت میوههای لیموترش خوشهای، اورکا و لیسبون بر تولید فلاونوئید هسپریدین میباشد. این تحقیق در قالب آزمایش فاکتوریل بر پایه طرح کاملاً تصادفی با سه عامل بافت (برونبر، میانبر و درونبر)، زمان برداشت (45، 75، 105 و 135 روز بعد از گلدهی کامل) و رقم (اورکا، لیسبون و خوشهای) در 3 تکرار انجام گرفت. متغییرهای اندازهگیری شده شامل وزن تر میوه، قطر میوه، ضخامت پوست میوه، درصد ماده خشک و مقدار هسپریدین موجود در عصاره بافتهای مختلف میوه بوده است. استخراج مواد با استفاده از متانول و اندازهگیری فلاونوئید با دستگاه HPLC صورت گرفت. نتایج این تحقیق نشان داد که وزن میوه، قطر میوه، ضخامت پوست میوه، مقادیر ماده خشک میوه و همچنین مقدار هسپریدین به شدت تحت تاثیر زمان برداشت میوه بوده است. تغییرات قطر میوه متناسب با وزن آن بوده است. بیشترین میزان وزن میوه در ارقام لیموترش لیسبون و لیموترش اورکا به ترتیب (39/88 و 72/84 گرم) در مرحله چهارم برداشت (135 روز پس از گلدهی کامل) و کمترین آن در لیموترش خوشهای (16/4 گرم) در مرحله اول برداشت (45 روز پس از گلدهی کامل) تولید شد. بیشترین میزان قطر میوه در رقم لیسبون (81/54 میلیمتر) در مرحله چهارم برداشت وکمترین قطر میوه (75/20 میلیمتر) در مرحله اول برداشت در رقم اورکا به وجود آمد. ضخامت پوست میوه ابتدا افزایش سپس کاهش یافته است. در بین ارقام، لیموترش خوشهای بیشترین ضخامت پوست میوه (16/7 میلیمتر) و لیموترش اورکا کمترین (66/4 میلیمتر) ضخامت پوست را داشته است. درصد ماده خشک میوهها در اولین مرحله برداشت افزایش، سپس کاهش یافت. بیشترین درصد وزن خشک در لیموترش خوشهای (83/22درصد) و کمترین آن در لیموترش لیسبون (36/11 درصد) تولید شدهاست. بیشترین میزان هسپریدین (93/1606 میلیگرم در لیتر) در لیموترش لیسبون، 45 روز بعد ازگلدهی کامل به دست آمد و بعد از آن تا زمان برداشت کاهش یافت. بیشترین میزان هسپریدین در بافت میانبر میوه تولید گردید. لیموترش لیسبون بهترین رقم لیمو از لحاظ مقدار هسپریدین در مقایسه با دو رقم دیگر میباشد.

تاثیر موقعیت فلس و نوع بستر کشت در تولید پیازچه به روش فلس جفتی در گیاه آماریلیس (Hippeastrum × johnsonii)
گیاه آماریلیس به عنوان گیاه زینتی گلدانی، فضای باز و گل شاخه بریده پرورش داده میشود. در شرایط عادی تعداد پیازچههای دختری تولید شده، بسیار کم میباشد. بنابراین بهبود روشهای تکثیر سنتی این گیاه، یکی از راهکارهای مناسب جهت کوتاه کردن دوره رشدی این گیاه میباشد. لذا این آزمایش با هدف بررسی اثر نوع بستر کشت و موقعیت قلمههای فلسی در پیاز مادری بر تولید پیازچه در شرایط آزمایشگاهی، به منظور افزایش سرعت تکثیر این گیاه زینتی، به صورت فاکتوریل در قالب طرح کاملا تصادفی با 7 تکرار انجام شد. جهت اندازهگیری صفات وزن تر و خشک از 3 تکرار استفاده گردید. پیازها به صورت شعاعی به 12 قطعه مساوی برش داده شدند، بطوری که هر قطعه دارای بخشی از صفحه پایگاهی باشد. به منظور بررسی اثر موقعیت فلس جفتی در پیاز مادری، قطعات برش خورده به 5 نمونه فلس جفتی تقسیم و گروه بندی شدند، بطوریکه در گروه یک، خارجیترین نمونههای فلس جفتی و در گروه پنج، داخلیترین نمونههای فلس جفتی قرار گرفتند. بسترهای کشت مورد استفاده در این آزمایش شامل بستر کشت ماسه، پرلایت، ورمیکولایت، پیت ماوس و کوکوپیت بود. در پایان آزمایش، تعداد پیازچه تولید شده در هر نمونه، قطر پیازچه، تعداد ریشه، طول ریشه، وزن تر و خشک اندامهای مختلف گیاه و میزان قهوهای شدن قلمههای فلسی ثبت گردید. نتایج نشان داد که بستر کشت و موقعیت فلس جفتی در پیاز مادری تاثیر معنیداری بر کیفیت پیازچههای باززایی شده داشتند. بیشترین میزان وزن تر پیازچه (58/1 گرم)، وزن خشک پیازچه (21/0 گرم) و بیشترین قطر پیازچه باززایی شده (5/1 سانتی متر) در بستر کشت پیت ماوس و فلس جفتی گروه یک حاصل گردید. بر اساس نتایج بدست آمده چنانچه نمونههای فلس جفتی از فلسهای خارجیتر پیاز تهیه گردند، پیازچههایی با کیفیت مطلوبتری تولید خواهند نمود. بطورکلی جهت تکثیر آماریلیس از طریق نمونههای فلس جفتی، کاربرد فلسهای جفتی گروه یک (خارجیترین لایه فلس جفتی) و بستر کشت پیت ماوس توصیه میگردد.

به منظور بررسی مقدماتی ویژگیهای زراعی و مورفولوژیک 107 توده اسفناج ایرانی (100 توده موجود در بانک ژن و 7 توده دیگر)، پژوهشی در ایستگاه تحقیقات کشاورزی کبوترآباد اصفهان طی سالهای 1391و 1392 انجام شد. در سال اول پژوهش بر اساس ویژگیهای برگ (رنگ، چروکیدگی، طول و عرض)، درصد بوتههای نر و ویژگیهای بذر، 25 توده از میان تودهها انتخاب شدند. در سال دوم آزمایش، 25 توده انتخابی با استفاده از طرح آماری بلوکهای کامل تصادفی با سه تکرار از نظر ویژگیهای عملکرد تازه، عملکرد ماده خشک، طول وعرض برگ، تعداد برگ و مقدار نیترات مقایسه شدند. 35 درصد از تودههای اسفناج استفاده شده در این پژوهش دارای رنگ برگ کاملاً سبز بودند و درصد بوتههای نر از 32 درصد تا 60 درصد در تودههای مختلف نوسان داشت. بیشترین مقدار عملکرد تازه برگ را رقم ورامین 88 با مقدار 59/32 تن در هکتار تولید نمود. سه توده TN-69-153، TN-69-140 و Varamin- prickly از نظر عملکرد تازه و عملکرد ماده خشک جزو تودههای برتر تشخیص داده شدند. سه توده TN-69-58، TN-69-101 و TN-69-70 کمترین مقادیر نیترات را داشتند. نتایج این پژوهش نشان داد در میان 107 توده مطالعه شده، ویژگیهای متنوع و مفید فراوانی از قبیل عملکرد بالا و مقدار نیترات پایین قابل تشخیص و جداسازی است و از این ویژگیها می توان در زمینههای اصلاحی و زراعی استفاده کرد.

تأثیر محلول پاشی اوره، نیترات کلسیم و اسیدبوریک بر رشد و عملکرد خیار گلخانه ای رقم خسیب
به منظور بررسی تأثیر محلول پاشی برگی بر روی خیار گلخانهای رقم خسیب، آزمایشی به مدت 5 ماه (اسفند ماه 86 تا تیرماه 87) در مرکز آموزش فنی و حرفهای زیبادشت واقع در 15 کیلومتری شهرستان کرج به اجرا گذاشته شد. برای دستیابی به عملکرد و کیفیت بالای محصول، محلول پاشی برگی در داخل گلخانههای سبزی و صیفی ضروری به نظر می رسد. به همین دلیل آزمایشی به صورت کرتهای دو بار خرد شده در سه تکرارانجام شد که در آن اثرات کود اوره، نیترات کلسیم و اسیدبوریک به ترتیب در غلظت های (0 و 3)، (0 و 10) و (0 و 5/0) گرم در لیتر بر روی صفات کمی و کیفی خیار گلخانهای از جمله عملکرد کل میوه، عملکرد میوه درجه یک، تعداد کل میوه، درصد میوههای درجه یک، قند میوه، طول بوته، درصد ماده خشک برگ و نسبت وزن برگ خیار مطالعه گردید. نتایج نشان داد که از بین اثرات ساده محلول پاشی، تیمار نیترات کلسیم بیشترین تأثیر را روی اکثر صفات مورد نظر گذاشت. همچنین از بین اثرات متقابل دوگانه محلول پاشی تیمار U1B1(اسیدبوریک – اوره در غلظت های 5/0و 3 گرم در لیتر) بیشترین تأثیر را روی عملکرد میوه درجه یک گذاشت. و از میان اثرات متقابل سه گانه محلول پاشی، تنها تیمار U1C0B0 (اسیدبوریک - نیترات کلسیم- اوره در غلظت های 0، 0 و 3 گرم در لیتر) بیشترین تأثیر را روی عملکرد کل میوه، عملکرد میوه درجه یک و تعداد کل میوه بر جای گذاشت. نتایج حاصل حاکی از آن است که تیمار نیترات کلسیم با بیشترین تأثیر گذاری بر روی اکثر صفات مورد نظر در رتبه اول قرار داشته و سایر تیمارها از جمله تیمار U1C0B0 و U1B1 به ترتیب در رده های بعدی تأثیر گذاری قرار داشتند.

بررسی اثر ترینگزاپک اتیل بر افزایش مقاومت به تنش خشکی در چمن علف گندمی بیابانی (Agropyron desertorum)
خشکسالی یکی از مهم ترین تنشهای غیر زنده برای رشد چمن در سراسر دنیا میباشد. کاهش کیفیت چمن ناشی از تنش خشکی یکی از نگرانیهای عمده در مدیریت چمنها میباشد. یکی از روشهایی که باعث افزایش مقاومت به تنشهای زیستی و غیر زیستی در گیاهان میگردد، استفاده از تنظیم کنندههای رشد است. هدف از این مطالعه بررسی اثر تنظیم کننده ترینگزاپکاتیل بر افزایش مقاومت به تنش خشکی در چمن علف گندمی بیابانی میباشد. آزمایش به صورت فاکتوریل بر پایهی طرح کاملأ تصادفی در سه تکرار به اجرا درآمد. تیمارها شامل سه سطح تنظیم کنندهی رشد ترینگزاپکاتیل (صفر، 25/0و 5/0 کیلوگرم در هکتار) و دو سطح تنش خشکی (آبیاری و قطع آبیاری) بود. نتایج نشان داد ترینگزاپکاتیل و خشکی به شکلی معنیدار موجب کاهش ارتفاع، وزن تر و خشک اندام هوایی گردیده است، ارتفاع در غلظتهای 25/0 و 5/0 کیلوگرم درهکتار به ترتیب 20/19 و 90/26 درصد کاهش نشان داد. ترینگزاپکاتیل بر خلاف خشکی موجب بهبود کیفیت و عرض برگ شد. خشکی موجب کاهش معنیدار محتوی نسبی آب و کلروفیل، افزایش پرولین و نشت الکترولیت گردید. ترینگزاپکاتیل با افزایش محتوی نسبی آب، پرولین، کلروفیل و کاهش نشت الکترولیت در شرایط تنش خشکی موجب افزایش مقاومت به خشکی در علف گندمی گردید. در شرایط خشکی عمق نفوذ ریشه و عمق موثر ریشه افزایش پیدا کرد، در حالی که ترینگزاپکاتیل تأثیر معنیداری بر خصوصیات ریشه نداشت. نتایج مشخص کرد ترینگزاپک اتیل میتواند موجب افزایش مقاومت به خشکی در علف گندمی گردد.

تاثیر تنش خشکی بر کیفیت ظاهری، خصوصیات مرفولوژیکی و فیزیولوژیکی 3 گونه چمن بومی
توسعه فضای سبز شهری و چمنکاری از جمله نیازهای بشر امروز است که عامل محدودکننده در این امر کمبود آب میباشد. این کمبود بر توسعه، کیفیت و انتخاب نوع پوشش گیاهی تاثیر مستقیم میگذارد. از اینرو آزمایش فاکتوریل بر پایه طرح کاملاً تصادفی برای سنجش مقاومت گونههای چمن بومی و انتخاب گونه مناسب روی گونههای بومی Brumos tomentellus، Festuca rubra، F .arundinacea و رقم تجاری Super sport (شاهد) و سه سطح رطوبتی 85 درصد (شاهد)، 65 درصد و 45 درصد ظرفیت زراعی در مرکز تحقیقات و آموزش کشاورزی و منابع طبیعی خراسان رضوی در شرایط گلخانه انجام گرفت. بر اساس نتایج بدست آمده بیشترین طول شاخساره در تنش خشکی 65 درصد ظرفیت زراعی در گونه F. arundinaceae و کمترین مقدار در چمن سوپر اسپورت، همچنین صفت عرض برگ بیشترین و کمترین مقدار در شرایط تنش به ترتیب در گونه B. tomentellus و چمن سوپر اسپورت مشاهده شد. گونه F. arundinaceae تحت شرایط تنش خشکی بهترین کیفیت ظاهری و بیشترین میزان کلروفیل a و کل را به خود اختصاص داد. تحت تنش خشکی کمترین نشت یونی (66/25 درصد) در گونه F.rubra، بیشترین مقدار محتوای آب نسبی در گونه F. arundinaceae (80/77 درصد)، بیشترین میزان پرولین برگ در گونه B. tomentellus و کمترین مقدار در چمن سوپر اسپورت مشاهده شد. با توجه به نتایج بدست آمده میتوان گفت گونههای چمن بومی در شرایط تنش خشکی عملکرد و کیفیت بهتری از چمنهای وارداتی مانند سوپر اسپورت را دارا میباشند.
