بررسی اثر سالیسیلیک اسید در جلوگیری از آسیب سرما در گوجه فرنگی گیلاسی (Lycopersicun esculentum cv. Messina)
سرما یکی از عوامل محدود کننده در تولید گوجه فرنگی محسوب میشود. تحمل به سرما در طی انبارداری یکی از شاخصه های مهم نگهداری محصولات باغی به شمار میرود. در این راستا آزمایش فاکتوریل برپایه طرح کاملا تصادفی به منظور اثر تیمار سالیسیلیک اسید در دورههای پیش و پس از برداشت روی ماندگاری گوجه فرنگی گیلاسی در دمای پایین انجام شد. بوتهها یکبار در مرحله تشکیل میوه با آب مقطر و سالیسیلیک اسید 75/0 میلیمولار و بار دوم میوههای هر دو بوته در مرحله رسیده کامل با همان غلظتهای سالیسیلیک اسید تیمار شده (آب مقطر-آب مقطر، آب مقطر- سالیسیلیک اسید، سالیسیلیک اسید- آب مقطر، سالیسیلیک اسید- سالیسیلیک اسید) و به مدت 5 هفته در دمای 1 درجه سانتی گراد نگهداری شدند. نتایج بدست آمده نشان داد که اثر سالیسیلیک اسید در مرحله پیش از برداشت بر روی آسیب سرمازدگی معنیدار بود و میوه های شاهد و تیمار شده به ترتیب 66/16 و38/1درصد آسیب را داشتند. بیشترین میزان مواد جامد محلول کل درهفته پنجم و اسیدیته در هفته چهارم مربوط به ترکیب تیماری سالیسیلیک اسید-آب مقطر می باشد که نشان می دهد تیمار پیش از برداشت سالیسیلیک اسید ضمن افزایش خصوصیات کیفی میوه، میزان پرولین و کربوهیدرات و افزایش فعالیت گایاکول پراکسیداز موجب کاهش آسیب غشای سلول شد. بطور کلی تاثیر تیمار پیش از برداشت سالیسیلیک اسید در حفظ کیفیت و افزایش ماندگاری گوجه فرنگی گیلاسی محسوس تر از تیمار پس از برداشت بود ولی با این وجود میوه های شاهد بهترین طعم را نسبت به سایر تیمارها در هفته پنجم داشتند.

تاثیر تاریخ کاشت بر عملکرد و تجمع نیترات اسفناج (.Spinacia oleracea L) در شرایط آب و هوایی اصفهان
به منظور بررسی تاریخ کاشتهای مختلف بر عملکرد، اجزای عملکرد و تجمع نیترات در اسفناج، پژوهشی دو ساله (1394-1393) در مرکز تحقیقات و آموزش کشاورزی و منابع طبیعی استان اصفهان انجام شد. بر اساس نتایج تجزیه مرکب دادهها تاثیر سال بر صفات آزمایشی معنیدار نبود و تاریخ کاشت 15 شهریور (571GDD=) با عملکرد 43 تن در هکتار بهترین تاریخ کاشت اسفناج تشخیص داده شد. با تاخیر تاریخ کاشت از 15 شهریور تا اول آبان علاوه بر کاهش معنیدار عملکرد، مقدار تجمع نیترات در گیاه به طور خطی افزایش یافت. مقادیر تجمع نیترات در تاریخ کاشتهای 15 شهریور (571GDD=)، اول مهر (354GDD=)، 15 مهر (193GDD=) و اول آبان (84GDD=) به ترتیب برابر بود با 2675، 2898، 3189 و 3571 قسمت در میلیون وزن تر. در تاریخ کاشتهای مختلف، تجمع نیترات در دمبرگ دامنهای از 3513 تا 4680 قسمت در میلیون وزن تر و در پهنک برگ دامنهای از 2135 تا 3125 قسمت در میلیون وزن تر داشت. با توجه به نتایج این پژوهش رعایت نکردن تاریخ کاشت مناسب نه تنها عملکرد اسفناج را کاهش خواهد داد بلکه با افزایش تجمع نیترات (بیش از 2500 قسمت در میلیون وزن تر) باعث مخاطرا تی برای مصرف کنندگان خواهد شد.

بررسی اثر نوع خاکپوش پلیاتیلنی بر رشد و عملکرد سیبزمینی (Solanum tuberosum)
بهمنظور بررسی تأثیر نوع خاکپوش پلیاتیلنی بر میزان رشد، زودرسی و عملکرد سیبزمینی پژوهشی بهصورت آزمایش فاکتوریل در قالب طرح بلوک کامل تصادفی در سه تکرار انجام شد. عوامل مورد آزمایش شامل خاکپوش با پلاستیک پلیاتیلنی در پنج سطح شامل پلاستیک شفاف، پلاستیک سفید، پلاستیک سیاه، پلاستیک دولایه سفید و بدون خاکپوش و عامل رقم در دو سطح شامل دو رقم آگریا و سانته بود. در طول دوره رشد زمان لازم تا 80 درصد سبز شدن، تعداد روز تا غدهزایی، ارتفاع گیاهان در مرحله گلدهی، قطر و تعداد ساقه اصلی در بوته، سطح برگ و طول دوره رشد اندازهگیری به عمل آمد. پس از برداشت غدههای تولیدی به درجات مختلف تفکیکشده و درصد ماده خشک آنها اندازهگیری شد. با نتایج تجزیه واریانس آزمایش مشخص شد که اثرات رقم و نوع خاکپوش پلیاتیلنی در اغلب صفات رشد معنیدار میباشد. خاکپوش اثر معنیداری بر درصد ماده خشک غده نداشت. اثرات متقابل رقم و خاکپوش در زمان لازم تا 80 درصد سبز شدن، تعداد ساقه اصلی، درصد غدههای تولیدی در اندازههای مختلف و عملکرد کل معنیدار شد. در پوشش پلیاتیلنی شفاف در کوتاهترین زمان ممکن، 80 درصد سبز شدن ایجاد شد. همچنین در پلیاتیلن شفاف در کوتاهترین زمان ممکن و با تفاوتی معنیدار با سایر پوششها و نیز تیمار بدون خاکپوش، غدهزایی انجام شد. در هر دو رقم، خاکپوش شفاف ارتفاع بوته بیشتری نسبت به سایر خاکپوشها و تیمار بدون خاکپوش ایجاد کرد. واکنش رقم و نوع پوشش پلیاتیلنی در تعداد ساقه اصلی متفاوت بود و بیشترین میزان تولید ساقه با متوسط 22/4 عدد در رقم سانته و پوشش شفاف حاصل شد. پوششهای پلیاتیلنی اثرات قابلتوجهی بر تعداد روز تا برداشت و درصد تولید غده در اندازههای کوچک (کوچکتر از 35 میلیمتر و غیرقابل فروش)، اندازه متوسط (35 تا 55 میلیمتر) و خوراکی (بزرگتر از 55 میلیمتر) و نیز عملکرد کل داشتند، هرچند پاسخ دو رقم به نوع پوششها متفاوت بود. درمجموع با تعدیل اثرات مثبت و منفی و بازارپسندی و همچنین عملکرد کل، پوشش شفاف با راندمان تولید غده بیشتر و با بازارپسندی نسبتاً بالاتر در مقایسه با سایر پوششها در تولید محصول سیبزمینی از برتری نسبی برخوردار بود.

اثر رقم و تیمار تنظیم کننده رشد گیاهی بر باززایی درون شیشهای لیلیوم با استفاده از ریزنمونههای TCL
لیلیوم به عنوان چهارمین گل شاخه بریده دنیا مطرح است و روش کشت بافت، روش مناسبی را برای تکثیر این گیاه زینتی فراهم کرده است. در این پژوهش به منظور بررسی عوامل مؤثر بر باززایی لیلیوم از پیازکهای تولید شده در شرایط درون شیشهای دو آزمایش جداگانه انجام گرفت. در مرحله اول، اثر ریزنمونههای با لایه سلولی نازک (TCL) با ضخامتهای 1، 3 و 5 میلیمتر در محیط کشت پایه موراشیگ و اسکوگ (MS) حاوی ترکیبهای تنظیم کننده رشد گیاهی BA، 2ip و Kin با غلظت 1 میلیگرم در لیتر در ترکیب با 5/0 میلیگرم در لیتر NAA بررسی شد. نتایج این آزمایش نشان داد که در هر سه تیمارتنظیم کننده رشد گیاهی، بیشترین میزان اندام زایی در ریزنمونههایی با ضخامت 3 میلیمتر بدست آمد. در واقع نتایج برهمکنش اندازه ریزنمونه با تنظیم کننده رشد گیاهی موجود در محیط کشت نشان داد درصد باززایی در ریزنمونههای با ضخامت 3 میلیمتر بهتر افزایش داشته است. بیشترین تعداد برگ و وزن خشک گیاهچهها در محیط کشت حاوی BA مشاهده شد که نسبت به سایر تیمارها تفاوت معنیداری داشت. در آزمایش دوم به منظور بررسی اثر رقم و نوع تنظیم کننده رشد گیاهی سیتوکینین، ریزنمونههای TCL ارقام Robina، Donato، Nymph، Lessoto و Roxana با ضخامت 3 میلیمتر در محیط کشت MS حاوی ترکیبات تنظیم کننده رشد گیاهی مختلف (BA، Kin، 2ip و TDZ) با غلظت 1 میلیگرم در لیتر در ترکیب با 5/0 میلیگرم در لیتر NAA کشت شدند. نتایج نشان داد که رقم به عنوان یکی از عوامل تأثیرگذار بر اندام زایی بود. بیشترین تعداد ریشه، وزن خشک، تعداد پیازک و طول گیاهچه در رقم Roxanaمشاهده شد. بررسی درصد باززایی و شاخهزایی در ریزنمونهها نشان داد که ارقام و هورمونهای مختلف بر میزان باززایی و ریشهزایی مؤثر بوده و رقم Nymph در حضور TDZ نسبت به سایر ارقام و تنظیم کننده های رشد گیاهی کمترین باززایی و ریشهزایی را نشان داد. همچنین مقایسه وزن خشک در تیمارهای مختلف نشان داد که وزن خشک گیاهچههای رقم Roxana بطور معنیداری نسبت به سایر تیمارها افزایش یافته بود.

تأثیر فورکلرفنورون بر پیازچهزایی، ترکیبات فیتوشیمیایی و خواص آنتیاکسیدانی موسیر (Allium hirtifolium)
این مطالعه با هدف بررسی اثر پیش تیمار و محلولپاشی فورکلرفنورون روی پیازچهزایی، ترکیبات فیتوشیمیایی و خواص آنتیاکسیدانی پیاز موسیر اجرا گردید. پیش تیمار پیازها از طریق غوطهور سازی آنها در غلظتهای مختلف فورکلرفنورون (0، 5 و 10 میلیگرم در لیتر) به مدت 24 ساعت انجام شد، همچنین محلولپاشی با چهار غلظت فورکلرفنورون (0، 50، 100 و 150 میلیگرم در لیتر) دو، چهار و شش هفته پس از کاشت صورت گرفت. پیش تیمار و محلولپاشی با فورکلرفنورون بهطور معنیداری تعداد برگ در هر بوته، وزن تر و وزن خشک پیازها را نسبت به نمونه شاهد افزایش دادند، ولی تعداد پیازچههای تولیدی تحت تأثیر تیمارها تغییری نشان نداد. مقدار آلیسین پیازهای موسیر تحت تأثیر تیمارهای مورد بررسی تغییر معنیداری نداشت و بهطور متوسط مقدار آن 859/0 میلیگرم در هر گرم بافت تازه پیاز بود. تیمار با فورکلرفنورون سبب افزایش معنیدار مقدار فنل کل پیازها گردید و بیشترین مقدار آن (585/1 میلیگرم گالیک اسید در هر گرم بافت تازه پیاز) در پیش تیمار 5 میلیگرم در لیتر و محلولپاشی 100 میلیگرم در لیتر فورکلرفنورون بهدست آمد. فعالیتهای آنزیمهای کاتالاز، پراکسیداز و آسکوربات پراکسیداز تحت تأثیر تیمارهای مختلف فورکلرفنورون افزایش معنیداری نسبت به نمونه شاهد نشان دادند. افزایش مقدار فنل و فعالیت آنزیمهای آنتیاکسیدان سبب افزایش فعالیت آنتیاکسیدانی پیازهای موسیر تیمار شده گردید. بر اساس نتایج بهدست آمده، پیش تیمار 10 میلیگرم در لیتر همراه با محلولپاشی 100 میلیگرم در لیتر با فورکلرفنورون میتواند راهکار مناسبی برای افزایش عملکرد و کیفیت پیازهای موسیر از طریق افزایش وزن و خواص آنتیاکسیدانی آنها باشد.

بررسی تاثیرکاربرد اکسید نیتریک بر کاهش اثرات نامطلوب سرما بر میوه پرتقال واشنگتن ناول (Citrus sinensis L. cv. Washington Navel) طی انبارداری
پرتقال واشنگتن ناول یکی از مهمترین مرکبات در ایران است. میوه پرتقال واشنگتن ناول که در دمای زیر 5 درجه سانتیگراد نگه داشته شود، عارضه فرورفتگی در پوست را به شدت نشان می دهد و افزایش در شدت علائم سرمازدگی با دمای پایین تر و مدت زمان انبارداری طولانی تر، افزایش می یابد. این آزمایش به منظور بررسی اثر اکسید نیتریک ، برکاهش خسارت سرمازدگی و ویژگیهای بیوشیمیایی پرتقال رقم واشنگتن ناول طی انبارداری انجام شده است. میوههای این رقم تجاری با غلظتهای صفر (شاهد)، 25/0 و 5/0 میلی مولار اکسید نیتریک تیمار و سپس در دمای 1±5 درجه سانتیگراد و رطوبت نسبی 90- 85 درصد، به مدت 5 ماه نگهداری شدند و ویژگیهایی نظیر سرمازدگی، نشت یون، پراکسیداسیون لیپیدها، پراکسید هیدروژن، مواد جامد محلول، اسیدهای آلی، pH، اسیدآسکوربیک و فعالیت آنزیمهای آنتیاکسیدانی (پراکسیداز و کاتالاز) طی 5 ماه انبارداری مورد ارزیابی قرار گرفتند. نتایج نشان داد، کاربرد 5/0 میلی مولار اکسید نیتریک نسبت به شاهد، به طور معنیداری خسارت سرمازدگی، نشت یون، پراکسیداسیون لیپیدها و پراکسید هیدروژن را کاهش و فعالیت آنزیم های آنتی اکسیدانی را افزایش داد. طبق این نتایج، مواد جامد محلول کل، pH، اسیدآسکوربیک میوههای تیمار شده و شاهد در طی دوره انبارداری روند افزایشی و میزان اسیدهای آلی روند کاهشی داشتند، اما استفاده از تیمار اکسید نیتریک، تغییرات دو روند مذکور را نسبت به شاهد کمتر تغییر داد. همچنین فعالیت آنزیمهای آنتی-اکسیدانی میوههای تیمار شده با اکسید نیتریک نسبت به شاهد، بسیار افزایش یافت. به طور کلی طبق نتایج حاصل از این پژوهش، میوههای تیمار شده با 5/0 میلی مولار اکسید نیتریک ، دارای بالاترین کیفیت و بیشترین فعالیت آنزیمهای آنتیاکسیدانی با کمترین خسارت سرمازدگی بودند.

مطالعه تأثیر کادمیم، روی و زئولیت بر خصوصیات فیزیومورفولوژیکی گل راعی (.Hypericum perforatum L)
در بین عناصر سنگین، کادمیم به دلیل تحرک و زیست فراهمی زیاد در خاک و ایجاد سمیت در غلظتهای کم، اهمیت ویژهای دارد. گل راعی، دومین گیاه پرمصرف در درمان افسردگی،به عنوان یک گیاه بیشاندوز کادمیم شناخته شده است. به دلیل ظرفیت تبادل کاتیونی بالای کانی زئولیت و شباهت رفتار شیمیایی یونهای روی و کادمیم، افزایش زئولیت و روی در محیط رشد گیاه، میتواند تعدیل کنندهی اثرات سمی کادمیم باشد. بدین منظور، آزمایشی به صورت فاکتوریل سه عاملی بر پایهی طرح بلوکهای کامل تصادفی با 27 تیمار و سه تکرار، به صورت گلدانی در مزرعه تحقیقاتی دانشکده کشاورزی دانشگاه فردوسی مشهد طی سالهای 1393-1392 اجرا گردید. تیمارها شامل سولفات کادمیم (0، 10، 20 میلیگرم در کیلوگرم خاک)، سولفات روی (0، 25، 50 میلیگرم در کیلوگرم خاک) و زئولیت معدنی (0، 5، 10 گرم در کیلوگرم خاک) بر خاک اعمال شدند. نتایج نشان داد که افزایش سطح کادمیم منجر به کاهش معنیدار صفات ارتفاع گیاه، طول ریشه، تعداد ساقه گلدهنده، تعداد گل، سطح برگ و میزان کلروفیل نسبی به ترتیب 28/6 ، 45/3 ، 04/2 ، 49/5 ، 82/0 و 71/15 درصد نسب به شاهد گردید و تنها، تعداد گرهکهای سیاه در برگ را 23/4 درصد نسبت به شاهد افزایش داد. تیمار ترکیبی روی و کادمیم تنها در صفات ارتفاع گیاه و طول ریشه توانست به طور معنیداری اثرات کاهنده کادمیم را کاهش دهد. همچنین اثر متقابل کادمیم و زئولیت تنها برای صفات طول ریشه و تعداد گل معنیدار شد. به طور کلی، کاربرد سولفات روی و زئولیت تا حدودی اثرات سوء کادمیم بر صفات اندازهگیری شده را تعدیل کرد.

اثرمحلول پاشی برخی عناصر ریز مغذیدر قبل و بعد از گلدهی بر ویژگی های کمی وکیفی میوه انگور رقم"خوشناو"
یکی از دلایل کاهش عملکرد در تاکستان ها و نیز کاهش کیفیت میوههای انگور،عدم مصرف متعادل کود می باشد. هدف از این پژوهش، تعیین بهترین غلظت و زمان کاربرد کود فروزنیک (حاوی آهن، روی و منگنز) بر برخی ویژگی های کمی و کیفی انگور تازه خوری رقم خوشناو بود. آزمایش به صورت فاکتوریل در قالب طرح بلوکهای کامل تصادفی با چهار تکرار انجام شد که فاکتور اول سطوح مختلف کود (0،500،1000،1500،2000 میلیگرم در لیتر) و فاکتور دوم زمان محلول پاشی (قبل از گلدهی، بعد از گلدهی و قبل و بعد از گلدهی) بود.نتایج نشان داد که اثر کاربرد کود فروزینک بر صفات متوسط وزن خوشه، متوسط وزن میوه در یک خوشه، نسبت TSS/TA، pH و ویتامین ث، معنیدار بود (p≤0.01). همچنین اثر زمان محلول پاشی بر تمام صفات به استثناء واکنش آب میوه معنیداری بود (p≤0.01). بیشترین وزن خوشه، متوسط وزن میوه در یک خوشه، TSS/TA،pH و ویتامین ث در تیمار کود (حاوی آهن، روی و منگنز) با غلظت 1000 میلیگرم در لیتر و در تیمار زمان گلدهی هر دو (قبل و بعد از گلدهی) به دست آمد.

با عنایت به کاهش نزولات جوی و پراکنش نامناسب بارندگی در طول سال، پدیده خشکی، یکی از معضلات فضای سبز شهری به شمار میرود. در واقع تنش خشکی یکی از مهمترین تنشهای محیطی میباشد که در مراحل مختلف رشد و نمو گیاه را تحت تأثیر قرار میدهد. بهمنظور بررسی تأثیر سطوح مختلف کمآبی بر سیستم آنتیاکسیدان و پراکسیداسیون چربی در گل جعفری، آزمایشی بهصورت فاکتوریل در قالب طرح پایه بلوکهای کامل تصادفی با 3 تکرار در دانشگاه شهید چمران اهواز در سال 1393 به اجرا درآمد. تیمارهای آزمایش شامل آبیاری در 3 سطح ETcrop 100 درصد (بدون تنش)، ETc75 درصد (تنش متوسط) و ETc50 درصد (تنش شدید) و ارقام گل جعفری آفریقایی و فرانسوی بود؛ در این پژوهش فعالیت آنزیمهای آنتیاکسیدانی (کاتالاز، پراکسیداز، آسکوربات پراکسیداز ) ، غلظت مالون دی آلدهید و رنگیزه های فتوسنتزی، 63 روز پس از اعمال تیمار اندازهگیری شدند؛ که طبق نتایج: تیمار آبیاری تاثیر معناداری بر کلروفیل a، b ، کل، شاخص محتوی کلروفیل ، فعالیت آنزیمهای کاتالاز، آسکوربات پراکسیداز، پراکسیداز و غلظت مالون دی آلدهید داشت. در حالیکه تفاوت معناداری بین دو گونه از گل جعفری بر هر یک از شاخص های اندازه گیری شده وجود نداشت. همچنین نتایج نشان داد میزان کلروفیل aو کلروفیل کل با افزایش تنش خشکی کاهش یافت، نتایج فعالیت آنزیمی نیز مشابه رنگیزه های فتوسنتزی و شاخص محتوی کلروفیل تحت تاثیر تنش خشکی، کاهش نشان داد. در کل طبق این پژوهش بین سطوح مختلف تیمار آبیاری از نظر میزان فعالیت آنزیمهای کاتالاز، آسکوربات پراکسیداز، غلظت مالون دی آلدهید، کلروفیل a، کلروفیل کل و غلظت کلروفیل تفاوت معنادار آماری وجود داشت بهگونهای که با کاهش مقدار آبیاری بر فعالیت آنزیم کاتالاز، آسکوربات پراکسیداز و غلظت مالون دی آلدهید افزوده شد و میزان رنگیزههای فتوسنتزی کاهش یافت. هرچند که تفاوتی بین دو گونه گل جعفری و اثر متقابل بین گونه و سطوح آبیاری از نظر شاخصهای اندازهگیری شده بهجز کلروفیل کل و کلروفیل b وجود نداشت.

اثر تنظیم کننده های رشد گیاهی بر کالوس زایی و باززایی لاله واژگون .Fritillaria imperialis L
لاله واژگون از زیباترین گلهای زینتی و داروئی بومی ایران است، که در معرض خطر انقراض میباشد. در این پژوهش کالوس های لاله واژگون به عنوان ریز نمونه در محیط کشت حاوی غلظت های مختلف NAA (0، 3/0 و 6/0 میلی گرم بر لیتر) در ترکیب با سه سایتوکنین BA (0، 3/0، 5/0 و 1 میلی گرم بر لیتر )، TDZ (0، 1/0، 3/0 و 5/0 میلی گرم بر لیتر) و Kin (0، 5/0، 1 و 5/1 میلی گرم بر لیتر) در قالب سه آزمایش جداگانه بر پایه طرح کاملا تصادفی کشت گردیدند. نتایج به دست آمده نشان داد که در هر سه آزمایش، در محیط کشت های حاوی 6/0 میلی گرم بر لیتر NAA در ترکیب با همه غلظت های سایتوکینین ها بیشترین میزان کالوس زایی (100 درصد ) به دست آمد. همچنین نتایج نشان داد هر چند کالوسزایی در محیطهای فاقد NAA نیز انجام گرفت، اما به منظور دستیابی به حداکثر کالوسزایی حضور NAA ضروری می باشد. بر اساس نتایج به دست آمده در محیط های فاقد سایتوکینین در هر سه آزمایش، باززایی صورت نپذیرفت. همچنین حضور NAA در ترکیب با Kin برای باززایی ضروری می باشد و در محیط هایی که تنها حاوی Kin می باشد، هیچ گونه باززایی مشاهده نشد.

تعیین زمان برداشت و عمر انبارمانی مناسب میوه نارنگی کینو در شهرستان جیرفت
مرکبات جزو میوههای نافرازگرا بوده و زمان برداشت و طول دوره انبارمانی میتواند تاثیر زیادی بر کیفیت میوه داشته باشد. این آزمایش به منظور بررسی خصوصیات کمی و کیفی میوههای برداشت شده نارنگی کینو در پانزدهم آذر، سیام آذر، پانزدهم دی، سیام دی و پانزدهم بهمن پس از 90-30 روز انبار خنک در دمای 6-4 درجه سانتیگراد در یکی از باغات شهرستان جیرفت اجرا گردید. نتایج نشان داد که برهمکنش زمان برداشت و مدت انبارمانی بر روی وزن میوه، گوشت، تفاله و عصاره و همچنین مواد جامد محلول (TSS)، اسید قابل تیتر (TA) و TSS/TA در سطح احتمال 1 درصد معنیدار گردید. تأخیر در برداشت باعث افزایش صفاتی از قبیل میزان وزن میوه، TSS و TSS/TA شد و بیشترین میزان TSS و TSS/TA مربوط به برداشت میوهها در 15 بهمن بود. با افزایش دوره انبارمانی، درصد کاهش وزن میوهها افزایش یافت. در مجموع، تاریخ برداشت پانزدهمام بهمن بهترین زمان برداشت جهت انبارداری میوه نارنگی کینو در جیرفت میباشد.

تعیین دماهای کاردینال چمن سردسیری و دو گونه علف هرز غالب آن
واکنش جوانهزنی بذر دو گونه چمن فتان قرمز(Festuca rubra L.) و چچم دائمی(Lolium perenne L.) و دو گونه علف هرز غالب آنها هفت بند (Polygonum aviculare L.) و شیر تیغی (Sonchus oleraceus L.) به دماهای 2، 5، 10، 15، 20، 25، 30، 35، 40 و 45 درجه سانتیگراد در ژرمیناتور مورد بررسی قرار گرفت. آزمایش در قالب طرح کاملا تصادفی و با 4 تکرار انجام شد. جهت تعیین دماهای کاردینال (پایه، مطلوب و بیشینه) از مدلهای رگرسیون غیر خطی (خطوط متقاطع، دندان مانند و چند جملهای درجه 2) بین سرعت جوانهزنی و دماهای مختلف استفاده شد. نتایج حاصل از تجزیه واریانس نشان داد که درصد جوانهزنی و سرعت جوانهزنی در سطح یک درصد تحت تاثیر دما قرار گرفتند. مدل خطوط متقاطع در گونه های چچم دائمی، فتان قرمز و شیرتیغی و مدل دندان مانند در گونه هفت بند به خوبی سرعت جوانهزنی را نسبت به دما توصیف نمودند و با توجه به کمترین جذر میانگین مربعات خطاء و بیشترین ضریب تبیین بهترین برازش را برای جوانهزنی بذور مورد بررسی نشان دادند. دمای پایه، مطلوب و بیشینیه به ترتیب برای چچم دائمی 12/4، 66/24، 27/43 برای فتان قرمز 0/2، 86/24، 48/43 برای هفت بند 95/2، 94/19- 21/22، 97/44 و برای شیرتیغی 0/2، 77/17، 86/44 درجه سانتیگراد به دست آمد. گونه فتان قرمز به علت پایینتر بودن دمای پایه نسبت به چچم دائمی و علف های هرز زودتر جوانه زد بنابراین استفاده از چمنهای اسپورتی که دارای نسبت بالاتری بذر فتان قرمز میباشند موجب میشود چمن بتواند سریعتر از گونههای هفت بند و شیرتیغی مستقر شده و در رقابت با علف های هرز پیروز باشد. با استفاده از نتایج این تحقیق میتوان کارایی کنترل علفهای هرز را در چمن بهبود بخشید.

بررسی تنوع ژنتیکی برخی ژنوتیپهای گردوی ایرانی در باغات تجاری مشهد با نشانگر ISSR
به منظور بررسی تنوع ژنتیکی تعدادی ژنوتیپهای گردوی ایرانی (Juglans regia L.) موجود در باغهای استان خراسان رضوی از نشانگر مولکولی ISSR استفاده گردید. با این هدف 56 ژنوتیپ گردو از باغهای استقلال، گلستان، الندشت و امام رضا انتخاب، پلاککوبی و مورد آزمایش واقع شدند. با استفاده از 9 آغازگر مورد استفاده، در مجموع 118 باند تولید شد که در محدوده 300 تا 3000 جفت باز بود و 29 باند چند شکل بودند. تعداد قطعات تکثیر شده برای هر آغازگر متفاوت بود، به طوری که آغازگر 844 تعداد 14 عدد و آغازگر 890 تعداد 8 عدد باند تکثیر شده داشت. متوسط قطعات تکثیر شده برای هر آغازگر 83/9 بود. در دندوگرام حاصل از خوشهبندی ژنوتیپها بر اساس ضریب تشابه دایس و روش UPGMA 10 کلاستر تشکیل شد. میزان تنوع ژنتیکی در جمعیت مورد مطالعه پایین بود که دلایل عمده آن دخالتهای انسانی، انتخاب و تکثیر تعداد معدودی از ژنوتیپهای برتر و گاها خود گرده افشانی گردو میباشد.

برآورد عملکرد بیومس گیاه مرزه تابستانه با استفاده از پارامترهای زودیافت خاک و شبکه عصبی مصنوعی
یکی از نیازهای مهم در برنامهریزی تولید و فرآوری گیاهان دارویی به منظور حصول عملکرد بالا و با کیفیت مطلوب، ارزیابی اولیه خصوصیات فیزیکی و شیمیایی خاک منطقه است که میتوان با اجتناب از کاربرد غیرضروری آزمایشات متنوع خاکشناسی، هزینه تولید را به حداقل کاهش داد. مرزه تابستانه (.Satureja hortensis L) ازجمله گیاهان دارویی پرکاربرد است که میزان اسانس و ترکیبات آن شاخص کیفی گیاه محسوب میشود. هدف از این پژوهش برآورد عملکرد بیومس گیاه مرزه تابستانه با استفاده از پارامترهای زودیافت خاک شامل: بافت خاک، موادآلی، pH، Ec، فسفر، پتاسیم، نیتروژن و درصد کربن با بهرهگیری از شبکهعصبیمصنوعی میباشد. بدین منظور 53 نمونه از خاک زراعی شهرستان نیشابور جمعآوری و پس از انجام آزمایشات ویژگیهای زودیافت خاک نتایج اولیه بدست آمد. بر پایه نتایج تجزیه حساسیت، کربن آلی، نیتروژن، فسفر، ماده آلی، پتاسیم، اسیدیته، شوری، رس، سیلت و شن به ترتیب به عنوان پارامترهای حساس در برآورد عملکرد بیومس بودند. درنهایت به منظور تخمین سریع و کم هزینه عملکرد بیومس، با استفاده از پارامترهای حساس، مدلهای گوناگون طراحی شدند. نتایج نهایی مقایسه مدلها نشان داد که بهترین مدل از نظر دقت و سرعت تخمینعملکرد بیومس مدلی بود که از ورودی با پارامتر بافت خاک و کربن آلی استفاده شد.

اثر کودهای سولفات آمونیوم و سولفات پتاسیم همراه با کود دامی بر شاخص های فیزیکوشیمیایی میوه و برگ خرما (.Phoenix dactylifera L) رقم مضافتی
با توجه به نیاز تغذیهای بالای درخت خرما (Phoenix dactylifera L.)، لازم است تا اثر کودهای پر مصرف بر شاخصهای فیزیکوشیمیایی برگ و میوه آن مورد بررسی قرار گیرد. برای این منظور، آزمایشی به صورت فاکتوریل در قالب طرح پایه بلوکهای کامل تصادفی شامل تیمار کودی سولفات آمونیوم و سولفات پتاسیم همراه با کود دامی به صورت چالکود در اسفند ماه و در منطقه بم اعمال گردید. نمونهبرداری جهت آنالیز برگ و میوه، به ترتیب در خرداد ماه و برداشت محصول خرما انجام شد. نتایج نشان داد با توجه به تاثیر معنیدار عناصر نیتروژن، گوگرد و پتاسیم فراهم شده توسط کودهای مذکور بر شاخصهای مورد نظر، بیشترین محتوای نیتروژن و آهن و رنگدانههای فتوسنتزی کاروتنویید و کلروفیل (a و کل) برگ و وزن میوه خرما به ترتیب از اثر متقابل کودهای سولفات آمونیوم و سولفات پتاسیم همراه با کود دامی، به دست آمد. بیشترین محتوای پتاسیم در برگ، TSS و نسبت TSS/TA در میوه، از تیمارهای سولفات آمونیوم (500 گرم) و سولفات پتاسیم (1500 گرم) همراه با کود دامی حاصل شد. بنابراین به منظور بهبود شاخصهای فیزیکوشیمیایی برگ و میوه درخت خرما، ترکیب کودی سولفات آمونیوم و سولفات پتاسیم همراه با کود دامی توصیه میشود.

غربالگری هموکاریون ها و تولیدنژاد دورگ جدید قارچ دکمهای سفید (Agaricusbisporus) با استفاده از نشانگر ریزماهواره
قارچ خوراکی دکمهای سفید (Agaricusbisporus) رایجترین قارچ خوراکی در ایران و دنیا است، اما بهدلایلی از جمله استفاده از نژادهای کم محصول وپسروی نژادی، عملکرد این قارچ درکشور کمتر از متوسط عملکرد آن در دنیا میباشد. یکی از برنامههای به نژادی،تولید نژاد F1 حاصل از آمیزش هموکاریونها است که مستلزم در اختیار داشتن جدایههای هموکاریون میباشد. در این مطالعه ابتدا 160جدایه تکاسپور کند رشد از نژاد A15 جداسازی شد و سپس براساس ویژگیهای مورفولوژیک، از بین آنها 18 نمونه بررسی و انتخاب شدند. در مرحلهی بعد، از نشانگر همبارز ریزماهواره (SSR) جهت شناسایی هموکاریونها استفاده شد. ده آغازگر که در نمونههای شاهد مادری دارای چندشکلی بودند برای نمونههای مورد آزمایش نیز استفاده شدند. جدایهها براساس حضور و عدم حضور باندهای چندشکل در دو گروه کلی هتروآللیک و هموآللیک قرار گرفتند و هفت جدایه که در تمامی جایگاههای بررسی شده الگوی باندی تک باند نشان دادند به عنوان جدایهی هموکاریون در نظر گرفته شدند. به منظور محاسبه سطح تنوع ژنتیکی بین 7 جدایه هموکاریون به دست آمده، ماتریس فاصله ژنتیکی با استفاده از نرمافزار NTSYSpcمحاسبه شد و دندروگرام مربوطه ترسیم گردید. تشابه ژنتیکی جفت نمونهها بین 17/0 تا 67/0متغیربود. در مرحلهی بعد نمونههای شماره 4 و 8 با بیشترین فاصله ژنتیکی نسبت به سایر نمونهها جهت تشکیل هیبرید در تلاقی شرکت داده شدند. نتیجه این تلاقی، تولید هیبرید N1 بود که با افزایش ناگهانی در رشد و تولید میسلیوم هوایی در محل الحاق همراه بود که در مقایسه با والدهای هموکاریون، سرعت رشد بیشتری داشت. در ادامه، جهت تأیید مولکولی هیبرید حاصل، واکنش
PCR-SSR با استفاده از یک پرایمر (AbSSR 45) انجام گرفت. همانطور که انتظار میرفت در هیبرید N1 همانند نمونهی شاهد هتروکاریون، دو باند قابل امتیازدهی حاصل شد که نشاندهندهی وجود دو هستهی غیرخواهری در هر واحد سلولی آن میباشد. نتایج نشان دادند که با استفاده از نشانگر SSR میتوان با احتمالی بیش از 8/99 درصد نسبت به قطعیت هموکاریونی پس از اجرای آزمایش با 10 آغازگر دست یافت و به این ترتیب از آنها در برنامههای اصلاحی جهت تولید هیبرید بهره برد.

ارزیابی تنوع ژنتیکی اکوتیپهای کلکسیون عناب ایران (Ziziphus spp.) با استفاده از نشانگر مولکولی ISSR
عناب (Zizyphus jujuba Mill.) به عنوان یک میوه ارزشمند و مقاوم به شرایط مختلف آب و هوایی در بسیاری از مناطق ایران گسترش دارد. از عناب علاوه بر صادرات بعنوان گیاه دارویی نیز استفاده میشود لذا ارزش اقتصادی قابل توجهی دارد. ارزیابی تنوع ژنتیکی در مواد گیاهی از اهمیت بالایی برخوردار بوده و گام اولیه برای شناسایی و نگهداری ذخایر ژنتیکی و از اصول اولیه اصلاح گیاهان محسوب میشود. استفاده از نشانگرهای DNA از بهترین روشهای موجود برای بررسی تنوع ژنتیکی میباشند. در این بررسی از نشانگر ISSR به منظور بررسی تنوع ژنتیکی 31 اکوتیپ عناب جمعآوری شده از هشت استان عنابخیز کشور استفاده گردید. تعداد 13 آغازگر در واکنش PCR آزمایش شدند که شش آغازگر DNAی الگو را بخوبی تکثیر و در بین نمونهها چندشکلی را آشکار نمودند. از میان 84 مکان ژنی شناسایی شده، تعداد 70 مکان (83 درصد) چندشکلی نشان دادند. تجزیه خوشهای اکوتیپها بر اساس ضریب تشابه دایس و به روش UPGMA انجام گرفت. در تجزیه خوشهای، نمونهها در سطح تشابه 85/0 در هفت گروه اصلی جای گرفتند. بیشترین تشابه ژنتیکی (95/0) بین نمونههای کلاله و درح و همچنین نوغاب و دوستیران و کمترین تشابه ژنتیکی (48/0) بین نمونههای برزادران و کنگان بدست آمد. آغازگرهای(AC)8YT و (GA)8Aمناسبترین آغازگرها برای مطالعات بعدی تشخیص داده شد. نتایج این مطالعه نشان داد که تنوع کل نمونههای کشور تقریباً در تنوع ناحیه خراسانجنوبی خلاصه شده است. همچنین در اکوتیپهای متعلق به استانهای اصفهان و مازندران، سطح قابل ملاحظهای از تنوع مشاهده شد.

بهینهسازی عوامل موثر در ازدیاد درون شیشهای پایه میروبالان c29
این مطالعه با هدف بررسی اثر محیط کشت و تنظیم کنندههای رشد بر شاخهزایی، ریشهزایی و سازگاری میروبالانC29 انجام شد. در این مطالعه از آزمایش فاکتوریل بر پایه طرح کاملا تصادفی با 4 تکرار و هر تکرار شامل 5 نمونه استفاده شد. به منظور ضد عفونی نمونه های گیاهی از 2 تیمار، شامل اتانول 70 درصد در 1 دقیقه و هیپوکلریت سدیم 10 درصد در 10 دقیقه استفاده شد. نتایج نشان داد هیپوکلریت سدیم 10 درصد به مدت 10 دقیقه، با 4 درصد آلودگی ریز نمونه های سال جاری بهترین تیمار ضدعفونی بود. از دو نوع محیط کشت شامل MS و DKW و تنظیم کننده رشد بنزیل آمینوپورین در غلظت های 0، 1، 2، 3 و 4 میلی گرم در لیتر در مرحله پرآوری و ایندول بوتیریک اسید در غلظت های 0، 1، 2 و3 میلی گرم در لیتر در مرحله ریشه زایی استفاده شد. نتایج نشان داد که محیط کشت MS حاوی 2 میلی گرم در لیتر بنزیل آمینو پورین با میانگین پر آوری 16/6 شاخساره، و طول 51/2 سانتی متر بیش ترین شاخه زایی و بیش ترین درصد ریشه زایی در محیط MS حاوی 2 میلی گرم در لیتر IBA و طول ریشه به میزان 100 درصد و 75/6 سانتی متر، به دست آمد. سازگاری گیاهچه ها به شرایط گلخانه ای با استفاده از سیستم مه افشانی، با موفقیت انجام شد بالاترین درصد بقای گیاهچه (0/70 درصد) در محیط کوکوپیت و پرلایت (1:1 حجمی) به دست آمد.
