بررسی تاثیر برخی محیط‌ کشت‌های پایه و تنظیم‌کننده‌های رشد گیاهی مختلف بر رشد درون شیشه‌ای فندق

چکیده

این تحقیق به منظور بررسی تاثیر چند نوع محیط کشت پایه و تنظیم‌کننده‌های رشد گیاهی بر استقرار و پرآوری درون‌شیشه‌ای فندق (Corylus avellana) انجام گرفت. برای این منظور، جوانه‌ها‌ی جانبی و انتهایی رقم فرتیل فندق در فصل بهار تهیه شده و پس از ضدعفونی روی محیط کشت‌های NRM، MS و MS½ حاوی 01/0 میلی‌گرم در لیتر IBA و سطوح مختلف BAP کشت شدند. نتایج نشان داد که بین محیط کشت‌های مختلف و همچنین سطوح متفاوت BAP تفاوت معنی‌داری از نظر درصد شاخه‌‌دهی، تعداد برگ در هر ریزنمونه و طول ساقه رشد کرده وجود دارد. بر‌اساس مقایسات گروهی مستقل، اگرچه درصد شاخه‌دهی محیط کشت پایه MS به طور معنی‌داری بیشتر از محیط کشت NRM و MS½ بوده ولی طول ساقه‌های رشد کرده در محیط کشت NRM به طور معنی‌داری بیشتر از دو محیط کشت پایه دیگر بود. به‌طوری که محیط کشت NRM حاوی 01/0 میلی‌گرم در لیتر IBA و یک میلی‌گرم بر لیتر BAP با 50% شاخه‌دهی، 33/5 برگ در هر ساقه و cm 6/1 طول ساقه رشد کرده مناسبترین شرایط را برای مرحله استقرار و رشد ریزنمونه‌ها فراهم آورد. ساقه‌های رشد کرده در مرحله استقرار، به صورت ریزنمونه‌های تک گره به محیط کشت NRM حاوی 05/0 میلی‌گرم در لیتر IBA و غلظت‌های مختلف BAP و TDZ انتقال داده شد. بیشترین درصد رشد به همراه کمترین درصد کالوس‌زایی و قهوه‌ای شدن در تیمار 5 میلی‌گرم بر لیتر BAP به همراه 05/0 میلی‌گرم بر لیتر IBA به دست آمد.

In this research, the effects of different basal medium and plant growth regulators on in vitro establishment and growth of hazelnut were investigated. For this, the spring apical and auxiliary buds of cv. Fertile were sterilized and cultured on NRM, MS and 1/2MS basal media containing 0.01 mg/l IBA and different levels of BAP. The results indicated that percentage of explant growth (shooting), number of leaves per explant and shoot length influenced significantly by basal media and concentration of plant growth regulator. Based on orthogonal contrasts analysis, although the highest percentage of shooting was obtained on MS medium, shoot length of explants cultured on NRM basal medium were significantly higher than those of MS and 1/2MS. The best growth response of explants in establishment stage (50% shooting, 5.33 leaves per shoot and 1.6 cm shoot length) were obtained with NRM medium supplemented with 0.01 mg/l IBA and 1 mg/l BAP. Shoots derived from establishment stage were cut to single-node explants and transferred on NRM medium supplemented with 0.05 mg/l IBA and different levels of BAP and TDZ. The highest percentage of explants growth with lowest callgenesis and explnt browning were obtained on NRM medium containing 0.05 mg/l IBA and 5 mg/l BAP.
