چکیده

خرفه، Portulaca oleracea، به علت داشتن ترکیبات آنتی‌اکسیدانی فراوان و اسیدهای چرب مهم مانند امگا-3 و 6، به‌عنوان گیاهی ارزشمند مورد توجه می‌باشد. در آزمایش اول ریزنمونه‌های حاصل از دانهال‌های استریل (برگ، ساقه و جوانه انتهایی) در محیط‌کشت‌های MS و ½MS حاوی غلظت‌های 0، 1/0، 3/0 و 5/0 میلی‌گرم در لیتر BAP و NAA در قالب طرح کاملاً تصادفی به منظور توان پینه‌زایی مورد بررسی قرار گرفتند. صفات مورفولوژیکی پینه‌ها پس از 5 ماه رشد در دمای 25 درجه سانتی‌گراد و 16/8 ساعت به ترتیب روشنایی و تاریکی، بررسی شد. مشاهدات مؤید تشکیل پینه‌های بادوام، سبزرنگ و دارای بافت فشرده‌ از ریزنمونه‌های ساقه بودند. بیشترین درصد پینه‌زایی، اندازه، وزن‌تر و خشک پینه به محیط‌کشت MS حاوی غلظت توأم 5/0 میلی‌گرم در لیتر BAP و NAA تعلق داشت. آزمایش دوم با هدف بررسی عصاره مخمر بر محتوای ترکیبات فنلی و ظرفیت آنتی‌اکسیدانی پینه‌های حاصل انجام‌ شد. برای این منظور پینه‌های 21 روزه به محیط‌کشت  MSدارای تیمار هورمونی منتخب آزمایش اول منتقل و با غلظت‌های صفر، 125، 250 و 500 میلی‌گرم در لیتر عصاره مخمر تیمار شدند. بیشترین محتوای فنل (12/664 میلی‌گرم اسید گالیک در صد گرم وزن خشک)، فلاونوئید (25/42 میلی‌گرم کوئرستین در صد گرم وزن خشک) و FRAP (0/787 میکرو مول آهن در گرم وزن خشک) به پینه‌های تیمار شده با 500 میلی‌گرم در لیتر عصاره مخمر تعلق داشت. در این آزمایش حداکثر میزان DPPH IC50 (45/2 میلی‌گرم در میلی‌لیتر) به تیمار شاهد اختصاص داشت.
