Tissue culture techniques offer suitable method for micropropagation of this ornamental plant. However, one of the problems during in vitro culture of carnation is vitrification. Ratio of ammonium to nitrate and agar concentrations in the medium affect this phenomenon. Therefore, in this study the effect of these factors on the rate of proliferation and the vitrification of carnation (Dianthus caryophyllus L.) cultivar Innove Orange Bogr, was evaluated. In this investigation lateral buds were cultured on MS medium containing 1 mg/l BA, 0.1 mg/l NAA and with different concentrations of agar and different ratio of ammonium to nitrate. The results showed that increasing in the agar concentration to 12 g/l lead to decreasing the rate of vitrification but regeneration also declined. Increase of agar concentration cause limitation in nutrient absorption by plants. Also, decrease in the ratio of ammonium to nitrate in the medium reduces the amount of vitrification, but did not result in adverse effects on plant regeneration rates. Multiple regressions showed that the effect of ammonium to nitrate ratio on vitrification was higher than agar concentration. So by considering the amount of shoot regeneration and vitrification, to obtain the most normal shoots, the concentration of 10 g/l agar with ammonium to nitrate ratio 1:6, is recommended.}, keywords = {Carnation,Ammonium to nitrate ratio,Agar concentration,Vitrification,Proliferation}, title_fa = {بررسی اثر نسبت آمونیوم به نیترات و غلظت آگار بر ریزازدیادی میخک (Dianthus caryophyllus L.) رقم Mondeo Kgr}, abstract_fa = {میخک به عنوان سومین گل شاخه بریده دنیا مطرح است و تکنیک‌های کشت بافت روش مناسبی را برای تکثیر این گیاه زینتی فراهم کرده است. با این وجود، یکی از مشکلات کشت درون شیشه‌ای میخک پدیده‌ی شیشه‌ای شدن می‌باشد. نسبت آمونیوم به نیترات و غلظت آگار در محیط کشت از عوامل موثر بر این پدیده می‌باشند. لذا این آزمایش به‌ منظور بررسی تاثیر این دو عامل بر میزان تکثیر و پدیده‌ی شیشه‌ای شدن میخک رقمMondeo Kgr، صورت گرفت. در این بررسی، ریزنمونه‌های جوانه‌ی جانبی در محیط کشت MS حاوی 1 میلی‌گرم در لیتر BA و 1/0 میلی‌گرم در لیتر NAA همراه با تیمارهای مختلف (غلظت‌های متفاوت آگار و نسبت‌های مختلف نیترات به آمونیوم) کشت گردیدند. نتایج آزمایش نشان داد که با افزایش غلظت آگار تا 12 گرم در لیتر، درصد شیشه‌ای شدن کاهش می‌یابد. در عین حال میزان باززایی ریزنمونه‌ها نیز کاهش می‌یابد، زیرا افزایش غلظت آگار باعث محدودیت در جذب مواد غذایی توسط گیاه می‌شود. همچنین با کاهش نسبت آمونیوم به نیترات میزان پدیده شیشه‌ای شدن کاهش یافت، ولی اثر نامطلوبی بر میزان باززایی گیاهچه‌ها نداشت. همچنین رگرسیون چند متغیره نشان داد که اثر نسبت آمونیوم به نیترات بسیار بیشتر از غلظت آگار در پدیده شیشه ای شدن موثر می‌باشد. لذا با درنظر گرفتن میزان شاخه‌زایی ریزنمونه‌های کشت شده و میزان شیشه‌ای شدن آنها، برای بدست آوردن بیشترین شاخه طبیعی، غلظت 10 گرم در لیتر آگار و نسبت 6:1 آمونیوم به نیترات توصیه می شود.}, keywords_fa = {میخک,نسبت آمونیوم به نیترات,غلظت آگار,شیشه ای شدن,پرآوری}, url = {https://jhs.um.ac.ir/article_30940.html}, eprint = {https://jhs.um.ac.ir/article_30940_6875a66c18a9e597fd3e21421e4b20f6.pdf} }
