تأثیر اتیل‌متان‌سولفونات (EMS) بر صفات مورفولوژیکی و فیتوشیمیایی و تغییرات ژنتیکی گیاه اسفناج (Spinacia oleracea L) رقم ویروفلای

چکیده
این پژوهش با هدف بررسی اثر غلظت‌ها و زمان‌های مختلف تیمار اتیل‌متان‌سولفونات (EMS) بر صفات مورفوفیزیولوژیکی و تغییرات ژنتیکی گیاه اسفناج (Spinacia oleracea L.) رقم ویروفلای انجام شد. آزمایش در سال‌های ۱۴۰۱-۱۴۰۲ در گلخانه علوم باغبانی دانشگاه کردستان به‌صورت فاکتوریل در قالب طرح کاملاً تصادفی اجرا گردید. بذرهای اسفناج با چهار غلظت EMS شامل صفر (شاهد)، 25/0، 5/0 و ۱ درصد و در دو زمان تیمار ۷۵ و ۱۵۰ دقیقه مورد تیمار قرار گرفتند. پس از شست‌وشوی کامل بذرها، کشت در شرایط یکنواخت گلخانه‌ای انجام شد و صفات مورفولوژیکی شامل ارتفاع بوته، وزن تر و وزن خشک شاخساره اندازه‌گیری گردید. همچنین برخی شاخص‌های فیتوشیمیایی از جمله میزان فلاونوئید، ویتامینC، ظرفیت آنتی‌اکسیدانی، نیترات، اسید اگزالیک و عناصر معدنی سدیم و پتاسیم مورد ارزیابی قرار گرفت. به‌منظور بررسی تغییرات ژنتیکی، از نشانگرهای مولکولی ISSR و SCOT برای تحلیل الگوی باندهای DNA استفاده شد.

نتایج نشان داد که با افزایش غلظت EMS، ارتفاع بوته به‌طور معنی‌داری کاهش یافت یعنی از 96/10 سانتی‌متر در تیمار شاهد به 66/8 سانتی‌متر در غلظت یک درصد رسید. در مقابل، غلظت‌های پایین EMS اثر مثبتی بر تجمع زیست‌توده داشت. بیشترین وزن تر شاخساره (56/15 گرم) و وزن خشک شاخساره (76/1 گرم) در غلظت 25/0 درصد مشاهده شد. شاخص‌های فیتوشیمیایی نیز تحت تأثیر EMS قرار گرفتند مثلاً در غلظت یک درصد بیشترین میزان آسکوربیک اسید ppm 709/1 مشاهده شد و همچنین بیشترین میزان فلاونوید (mg/gFW 17/13 ) و ظرفیت آنتی‌اکسیدانی (µmol/g FW 44/37 ) در غلظت 25/0 درصد EMS مشاهده شد. نتایج حاصل از تجزیه مولکولی نشان داد که تیمار EMS موجب تغییر در الگوی باندهای DNA، ظهور باندهای جدید و حذف برخی باندهای موجود گردید که بیانگر افزایش تنوع ژنتیکی در گیاهان شده است. به‌طور کلی، نتایج این پژوهش نشان می‌دهد که کاربرد کنترل‌شده EMS در غلظت‌های پایین می‌تواند روشی مؤثر برای القای جهش‌های مفید و افزایش تنوع ژنتیکی در اسفناج رقم ویروفلای باشد.

This research aimed to investigate the effect of different concentrations and treatment durations of Ethyl Methanesulfonate (EMS) on the morphophysiological traits and genetic variations of spinach (Spinacia oleracea L.) ‘Viroflay’ cultivar. The experiment was conducted during the years 2022-2023 in the horticultural greenhouse of the University of Kurdistan, using a factorial experiment in a completely randomized design. Spinach seeds were treated with four concentrations of EMS: 0 (control), 0.25%, 0.5%, and 1%, and exposed for two treatment times: 75 and 150 minutes. After thorough seed washing, cultivation was carried out under uniform greenhouse conditions. Morphological traits, including plant height, shoot fresh weight, and shoot dry weight, were measured. Furthermore, several phytochemical indices were evaluated, such as flavonoid content, Vitamin C, antioxidant capacity, nitrate, oxalic acid, and mineral elements (sodium and potassium). To assess genetic variations, ISSR and SCOT molecular markers were employed to analyze DNA banding patterns.

The results indicated that increasing EMS concentration significantly reduced plant height, decreasing from 10.96 cm in the control treatment to 8.66 cm at a 1% concentration. Conversely, low EMS concentrations had a positive effect on biomass accumulation. The highest shoot fresh weight (15.56 g) and shoot dry weight (1.76 g) were observed at the 0.25% EMS concentration. Phytochemical indices were also affected by EMS; for instance, the highest ascorbic acid content (709.17 ppm) was observed at the 1% concentration. Additionally, the highest flavonoid content (13.17 mg/g FW) and antioxidant capacity (37.44 µmol/g FW) were recorded at the 0.25% EMS concentration. Molecular analysis results revealed that EMS treatment altered DNA banding patterns, leading to the appearance of new bands and the disappearance of some existing ones, suggesting an increase in genetic diversity.
