بهبود تحمل تنش خشکی فلفل دلمه‌ای از طریق محلول‌پاشی ملاتونین و سیلیسیم

چکیده
فلفل دلمه‌ای (Capsicum annuum L.) یکی از سبزیجات مهم و پرمصرف در سبد غذایی انسان است که به دلیل ویژگی‌های فیزیولوژیکی خاص، نسبت به تنش خشکی و شرایط کم‌آبی بسیار حساس می‌باشد. در سال‌های اخیر، استفاده از ترکیبات زیستی به‌عنوان الیسیتورهای طبیعی برای افزایش مقاومت گیاهان در برابر تنش‌های غیرزیستی مورد توجه قرار گرفته است. این پژوهش با هدف بررسی اثر محلول‌پاشی برگی ملاتونین و سیلیسیم بر شاخص‌های فیزیولوژیک و دفاعی فلفل دلمه‌ای تحت تنش خشکی انجام شد. آزمایش به‌صورت اسپلیت فاکتوریل در قالب طرح کاملاً تصادفی با فاکتور اصلی خشکی در سه سطح (90، ۷۰ و 50 درصد ظرفیت زراعی)، فاکتور فرعی اول، ملاتونین در سه سطح (۰، ۱۰۰ و ۲۰۰ میکرومولار) و فاکتور فرعی دوم، سیلیسیم در سه سطح (۰، ۱۰۰ و ۲۰۰ میلی‌گرم در لیتر) در گلخانه دانشگاه علوم کشاورزی و منابع طبیعی ساری اجرا گردید. صفات مورد بررسی شامل فعالیت آنزیم‌های آنتی‌اکسیدانی (کاتالاز، پراکسیداز، سوپراکسید دیسموتاز)، میزان پرولین، مالون دی‌آلدئید (MDA) و کربوهیدرات‌های محلول برگ بود. نتایج تجزیه واریانس نشان داد که اثرات متقابل تیمارها بر میزان آنزیم‌‌‌های کاتالاز و پراکسیداز، کربوهیدرات محلول برگ، پرولین و مالون‌دی‌آلدئید در سطح احتمال یک درصد معنی‌دار بود. بیشترین میزان فعالیت آنزیم‌های کاتالاز و پراکسیداز در شرایط بدون تنش و محلول‌پاشی 100 میکرومولار ملاتونین و 100 میلی‌گرم در لیتر سیلیسیم بدست آمد. همچنین بیشترین تجمع پرولین در بالاترین سطح تنش همراه با محلول‌پاشی ملاتونین ۲۰۰ میکرومولار مشاهده شد. محلول‌پاشی با سیلیسیم در سطوح 100 و 200 میلی‌گرم در لیتر و 200 میکرومولار ملاتونین به طور معنی‌داری میزان MDA در بوته‌های تحت تنش را کاهش داد. بیشترین میزان کربوهیدرات محلول برگ در تیمار تنش متوسط و محلول‌پاشی سیلیسیم ۱۰۰ میلی‌گرم بر لیتر مشاهده گردید. این نتایج نشان می‌دهد که کاربرد همزمان ملاتونین و سیلیسیم با تقویت سیستم دفاعی آنتی‌اکسیدانی، افزایش تجمع اسمولیت‌ها و کاهش آسیب‌های اکسیداتیو، تحمل فلفل دلمه‌ای را در برابر تنش خشکی به‌طور مؤثری ارتقاء می‌دهد. بنابراین، استفاده تلفیقی از این دو ترکیب می‌تواند به‌عنوان راهکاری زیستی و کم‌خطر برای مدیریت تنش خشکی در تولید پایدار این گیاه پیشنهاد شود.

Introduction

Bell pepper (Capsicum annuum L.) is one of the most important and widely consumed vegetables in the human diet. Due to its specific physiological characteristics, it is very sensitive to drought stress and water deficit conditions. In recent years, the use of biological compounds as natural elicitors to increase plant resistance to abiotic stresses has received attention. This study aimed to investigate the effect of foliar spraying of melatonin and silicon on physiological and defense indices of bell pepper under drought stress.

Materials and Methods

The experiment was conducted as a split factorial experiment in a completely randomized design with drought as the main factor at three levels (90, 70, and 50 percent of field capacity), the first subfactor, melatonin at three levels (0, 100, and 200 µM), and the second subfactor, silicon at three levels (0, 100, and 200 mg/L) in the greenhouse of Sari Agricultural Sciences and Natural Resources University. The studied traits included antioxidant enzyme activity (catalase, peroxidase, superoxide dismutase), proline, malondialdehyde (MDA) and soluble leaf carbohydrates.

Results and Discussion

The results of analysis of variance showed that the interaction effects of treatments on the levels of catalase and peroxidase enzymes, leaf soluble carbohydrates, proline, and malondialdehyde were significant at the 1% probability level. The highest activity of catalase and peroxidase enzymes was obtained under non-stress conditions and foliar application of 100 μM melatonin and 100 mg/L silicon. Also, the highest proline accumulation was observed at the highest stress level along with foliar application of 200 μM melatonin. Foliar application of silicon at levels of 100 and 200 mg/L and 200 μM melatonin significantly reduced the amount of MDA in stressed plants. The highest amount of leaf soluble carbohydrates was observed in the moderate stress treatment and 100 mg/L silicon foliar application.

Conclusion

These results indicate that simultaneous application of melatonin and silicon effectively improves the tolerance of bell pepper to drought stress by strengthening the antioxidant defense system, increasing the accumulation of osmolytes, and reducing oxidative damage. Therefore, the combined use of these two compounds can be proposed as a biological and low-risk solution for managing drought stress in the sustainable production of this plant.
